捕食線蟲性真菌-少孢節(jié)叢孢菌凍干生物制劑的制備

【摘要】:通過在含有0.05%瓊脂粉的沙氏葡萄糖肉湯培養(yǎng)基中培養(yǎng)少孢節(jié)叢孢菌(Arthrobotrys oligospora)再轉(zhuǎn)接到玉米粒培養(yǎng)基的方法,對(duì)少孢節(jié)叢孢菌分生孢子進(jìn)行培養(yǎng)。在不同時(shí)間點(diǎn),將培養(yǎng)好的分生孢子從培養(yǎng)基上洗脫并進(jìn)行計(jì)數(shù),然后制成凍干制劑,并觀察其復(fù)蘇情況,從而為少孢節(jié)叢孢菌凍干制劑的臨床應(yīng)用提供基礎(chǔ)數(shù)據(jù)。結(jié)果顯示:通過對(duì)少孢節(jié)叢孢菌凍干制劑在萌發(fā)率、生長(zhǎng)率和捕食率三方面的復(fù)蘇情況觀察,少孢節(jié)叢孢菌凍干后的分生孢子在接種后4d,萌發(fā)率達(dá)到最大值79.5%,平均生長(zhǎng)率為3.4mm/d,加入蟲體后7d捕食率最高,為95.8%,平均每天捕食率為74.0%;少孢節(jié)叢孢菌凍干制劑在平均生長(zhǎng)率和捕食率方面與對(duì)照組相比,差異均不顯著(P0.1)。結(jié)果表明,凍干制劑材料易得,方法簡(jiǎn)單易行,復(fù)蘇效果良好,進(jìn)一步優(yōu)化后將具有產(chǎn)業(yè)化應(yīng)用的前景。
[Abstract]:Arthrobotrys oligospora was cultured in a broth medium containing 0.05% Agar powder. The method of transferring to the corn kernel medium. At different time points, the cultured conidia were eluted from the culture medium and counted, then the lyophilized preparation was prepared and its recovery was observed. The results showed that the recovery rate, growth rate and predation rate of lyophilized preparation were observed in three aspects: germination rate, growth rate and predation rate. The germination rate of the lyophilized conidia reached a maximum of 79.5 mm / d at 4 days after inoculation, and the average growth rate was 3.4 mm / d, and the predation rate was the highest (95.8%) at 7 days after inoculation. The average daily predation rate was 74.0%; Compared with the control group, the average growth rate and predation rate of the lyophilized preparation were not significantly different from those of the control group (P 0.1). The results showed that the freeze-dried preparation was easy to obtain, the method was simple and feasible, and the effect of resuscitation was good. Further optimization will have the prospect of industrial application.
