【摘要】：為探明2014年-2015年在福建省流行的羊傳染性膿皰病毒(ORFV)遺傳變異情況,對10株ORFV流行毒株的F1L、B2L和VIR基因進行克隆、測序及分析。結果表明,10株ORFV F1L基因之間的核苷酸序列同源性為97.6%~100%,與國內株的核苷酸序列同源性為96.8%~99.7%,與NZ2參考株的核苷酸序列同源性為96.3%~97.1%;同NZ2參考株比較,FJ-YT2014缺失2個氨基酸;10株ORFV B2L基因之間核苷酸序列同源性為97.5%~99.9%,與國內株的核苷酸序列同源性為96.7%~99.5%,與NZ2參考株的核苷酸序列同源性為96.7%~97.7%;10株ORFV VIR基因之間的核苷酸序列同源性為95.8%~99.5%,與國內株的核苷酸序列同源性為94.6%~99.6%,與NZ2參考株的核苷酸序列同源性為94.6%~96.4%�；诨蚝塑账嵝蛄械倪z傳進化分析表明,10株ORFV F1L基因與福建省分離株、山西株和新疆株親緣關系較近;10株ORFV B2L基因與新疆、山西、德國毒株親緣關系較近;10株ORFV VIR基因與臺灣、新疆株親緣關系較近。結果提示,當前福建省流行的ORFV F1L、B2L和VIR基因尚未出現(xiàn)明顯變異,但是其F1L、B2L和VIR基因核苷酸序列之間普遍存在異質性。
[Abstract]:In order to investigate the genetic variation of infectious pustular virus (ORFV) from 2014 to 2015 in Fujian Province, the F1L of 10 strains of ORFV were studied. B2L gene and VIR gene were cloned, sequenced and analyzed. The results showed that the nucleotide sequence homology between ORFV F1L gene and B2L gene was 97.6%. The homology of nucleotide sequence with domestic strain was 96.8and 99.70.The homology with NZ2 reference strain was 96.3and 97.1; Compared with NZ2 reference strain, FJ-YT2014 lacks 2 amino acids. The homology of nucleotide sequence between 10 strains of ORFV B2L gene and domestic strain was 97.5% and 99.9%, respectively. The nucleotide sequence homology with NZ2 reference strain was 96.7%. The nucleotide sequence homology among the 10 strains of ORFV VIR gene was 95.88.99. 5%, and the homology of nucleotide sequence with domestic strains was 94.6%. The nucleotide sequence homology with NZ2 reference strain was 94.6%. The genetic evolution analysis based on the nucleotide sequence showed that ORFV F1L gene of 10 strains and Fujian Province isolate. The relationship between Shanxi plant and Xinjiang plant is close; 10 strains of ORFV B2L gene were closely related to Xinjiang, Shanxi and Germany. 10 strains of ORFV VIR gene were closely related to those in Taiwan and Xinjiang. The results suggested that there was no significant variation in the ORFV F1L LU B2L and VIR genes in Fujian Province at present. But the nucleotide sequence of F 1 L B 2 L and VIR gene is heterogeneity.
