FGF2和FGFR在牦牛繁殖周期子宮的表達(dá)與定位

【摘要】:為探索成纖維細(xì)胞因子2(fibroblast growth factor,F2GF2)及成纖維細(xì)胞因子受體(fibroblast growth factor receptor,FGFR)在健康成年牦牛繁殖周期中的生物學(xué)功能,本研究選取了不同繁殖周期(發(fā)情期-卵泡期、發(fā)情期-黃體期、妊娠期-黃體期)牦牛子宮組織,采用實(shí)時(shí)熒光定量PCR(quantitative real-time PCR,RT-qPCR)和免疫組織化學(xué)方法從基因和蛋白水平分別檢測(cè)不同繁殖周期牦牛子宮FGF2和FGFR的表達(dá)和分布,用積分光密度方法分析FGF2和FGFR蛋白表達(dá)水平。結(jié)果顯示,不同繁殖周期牦牛子宮中FGF2mRNA表達(dá)水平為:發(fā)情期-卵泡期發(fā)情期-黃體期妊娠期-黃體期(P0.01);FGFRmRNA表達(dá)水平為:發(fā)情期-黃體期妊娠期-黃體期發(fā)情期-卵泡期(P0.05)。FGF2和FGFR在牦牛不同繁殖周期的子宮內(nèi)膜、子宮腺及血管上皮中均有表達(dá),子宮內(nèi)膜和子宮腺為主要表達(dá)部位,陽(yáng)性細(xì)胞的細(xì)胞質(zhì)中表達(dá)較高。積分光密度分析結(jié)果顯示,FGF2蛋白表達(dá)水平為:發(fā)情期-卵泡期子宮發(fā)情期-黃體期子宮妊娠期-黃體期子宮;FGFR蛋白表達(dá)水平為:妊娠期-黃體期子宮發(fā)情期-卵泡期子宮發(fā)情期-黃體期子宮。以上結(jié)果表明,FGF2和FGFR在不同繁殖周期的表達(dá)差異性可能與其參與早期胚胎附植及胚胎發(fā)育的調(diào)控相關(guān),這為進(jìn)一步研究FGF2和FGFR在牦牛繁殖周期中的調(diào)控作用提供了理論依據(jù)。
[Abstract]:To explore fibroblast growth factor. F2GF2) and fibroblast growth factor receptor. The biological function of FGFR in the reproductive cycle of healthy adult yaks. In this study, different reproductive cycles (estrus-follicle, estrus-luteal phase, gestational-luteal phase) were selected. Real-time fluorescence quantitative PCR(quantitative real-time PCR was used. RT-qPCR) and immunohistochemical method were used to detect the expression and distribution of FGF2 and FGFR in yak uterus from gene and protein levels, respectively. The expression levels of FGF2 and FGFR proteins were analyzed by integrated optical density method. The expression levels of FGF2mRNA in the uterus of yak in different reproductive cycles were as follows: from estrus to follicle to luteal phase pregnancy to luteal phase P0.01; The expression levels of FGFRmRNA in the endometrium of yaks in different reproductive cycles were as follows: from estrus to luteal phase pregnancy to luteal period to follicular stage. The positive cells were expressed in the cytoplasm of the positive cells. The results of integrated optical density analysis showed that the positive cells were expressed in both uterine gland and vascular epithelium, and the endometrial and uterine glands were the main expression sites. The expression levels of FGF2 protein were: estrus, follicle, estrus, luteal, pregnancy, corpus luteum; The expression level of FGFR protein was: pregnancy luteal phase uterine estrus follicle phase uterine estrus phase luteal phase uterus. The difference of FGF2 and FGFR expression in different reproductive cycles may be related to their involvement in the regulation of early embryo implantation and embryo development. This provides a theoretical basis for further study on the regulation of FGF2 and FGFR in yak reproductive cycle.
