壽光雞AANAT基因5′調(diào)控區(qū)的單核苷酸多態(tài)性及其與產(chǎn)蛋性狀的關(guān)聯(lián)分析

【摘要】:褪黑激素(melatonin,Mel)分泌增加可阻礙家禽生殖器官的發(fā)育,降低家禽產(chǎn)蛋量。5-羥色胺-N-乙;D(zhuǎn)移酶(arylalkylamine N-acetyltransferase,AANAT)是Mel合成過(guò)程中的限速酶。本實(shí)驗(yàn)旨在研究AANAT基因5'調(diào)控區(qū)多態(tài)性及其與壽光雞(Gallus domesticus'Shouguang')產(chǎn)蛋性能的關(guān)系,采用直接測(cè)序法檢測(cè)了AANAT基因5'調(diào)控區(qū)的SNPs,并將檢測(cè)到的SNPs與壽光雞的產(chǎn)蛋性狀進(jìn)行關(guān)聯(lián)分析。結(jié)果表明,AANAT基因5'控區(qū)共21個(gè)SNPs,其中10個(gè)低度多態(tài)、11個(gè)中度多態(tài)。3個(gè)中度多態(tài)位點(diǎn)對(duì)產(chǎn)蛋性能影響顯著(P0.05),其中-803位點(diǎn)能使開產(chǎn)日齡顯著提前,并能使前期(20~25周齡)產(chǎn)蛋數(shù)顯著增加;-561位點(diǎn)能使中期產(chǎn)蛋數(shù)顯著增加;-522位點(diǎn)能使后期(46~57周齡)產(chǎn)蛋數(shù)顯著增加;-561、-522位點(diǎn)能使產(chǎn)蛋總數(shù)顯著增加。綜上所述,AANAT基因5'調(diào)控區(qū)有多個(gè)SNPs對(duì)產(chǎn)蛋性狀影響顯著,可作為壽光雞產(chǎn)蛋性狀輔助選擇的有潛在利用價(jià)值的分子遺傳標(biāo)記。
[Abstract]:The increased secretion of melatonin (melatonin) may hinder the development of reproductive organs in poultry. Decreased egg production. 5-hydroxytryptamine N-acetyltransferase (Arylalkylamine N-acetyltransferase). Aanat is a rate-limiting enzyme in the process of Mel synthesis. The aim of this study was to study the polymorphism of 5 'regulatory region of AANAT gene and its association with Shouguang chicken (. The relationship between Gallus domesticus and egg laying performance. SNPs of 5 'regulatory region of AANAT gene were detected by direct sequencing, and the results showed that the detected SNPs was associated with egg laying traits of Shouguang chicken. There were 21 SNPs in the 5'control region of AANAT gene, of which 10 were low polymorphic, 11 were moderate polymorphic, and 3 moderate polymorphic loci had significant effect on egg laying performance (P0.05). The -803 locus could significantly advance the first day and increase the number of eggs at 20 ~ 25 weeks. -561 locus could significantly increase the number of eggs laid in the middle stage. -522 locus could significantly increase the number of eggs laid at the later age of 46 ~ 57 weeks. The results showed that the total number of eggs was significantly increased at -561 ~ 522 locus. In conclusion, there were more than one SNPs in the 5 '-regulatory region of the AANAT gene, which had a significant effect on the egg laying traits. It can be used as a potentially useful molecular genetic marker for assisted selection of egg-laying traits in Shouguang Chicken.
