【摘要】:[目的]探討紫錐菊多糖(EPS)在內毒素(LPS)損傷后IEC-6分泌IL-6m RNA的影響。[方法]采用Trizon試劑提取總RNA,經(jīng)RT-PCR擴增后,進行瓊脂糖凝膠電泳及圖像分析。[結果]當LPS刺激IEC-6細胞時,IL-6m RNA分泌增加。而EPS可以抑制這種作用,并且具有劑量依賴性,50μg/ml EPS可以部分抑制LPS刺激IEC-6產(chǎn)生的IL-6水平,而100、500μg/ml EPS隨著濃度的增加,其抑制IL-6的水平逐漸增加;將IEC-6用50、100、200、500μg/ml EPS預處理24 h,然后用LPS(10μg/ml)刺激1、4h,采用RT-PCR方法分析得,LPS誘導IL-6m RNA表達被EPS抑制且具有時間依賴性。[結論]證實了LPS作用于小腸上皮細胞后,其分泌的細胞因子IL-6m RNA產(chǎn)生增多,而EPS可以抑制LPS刺激細胞分泌的IL-6m RNA的產(chǎn)生從而起到腸道粘膜的保護作用,并且這種抑制作用具有濃度及時間依賴性。
[Abstract]:[Objective] to investigate the Echinacea polysaccharide (EPS), (LPS)] IL-6m RNA. Methods IEC-6 secretion after injury to extract total RNA using Trizon reagent, amplified by RT-PCR, agarose gel electrophoresis and image analysis. Results when LPS stimulated IEC-6, IL-6m and EPS increased secretion of RNA. Can inhibit this effect, and in a dose dependent manner. 50 g/ml EPS could inhibit LPS stimulated IEC-6 production level of IL-6, and 100500 g/ml EPS with the increase of concentration, the inhibition of IL-6 levels gradually increased; the IEC-6 with 50100200500 g/ml EPS pretreatment for 24 h, then LPS (10 g/ml) stimulated 1,4h analysis by using RT-PCR method, LPS IL-6m induced RNA expression was inhibited by EPS and a time dependent manner. Conclusion] confirmed the effect of LPS on intestinal epithelial cells, the secretion of cytokines of IL-6m increased RNA production, and EPS can inhibit LPS cell stimulation The production of the secreted IL-6m RNA plays a protective role in the intestinal mucosa, and this inhibitory effect is dependent on concentration and time.
