Fas系統(tǒng)在睪酮所誘導(dǎo)的小鼠骨髓巨噬細胞凋亡中的作用

細胞凋亡與組織器官的發(fā)育及分化、免疫細胞的發(fā)生及功能的維持、腫瘤的發(fā)生及轉(zhuǎn)歸等均有密切的關(guān)系,細胞以凋亡的方式主動“自殺”對維持組織的自身穩(wěn)定具有積極意義。巨噬細胞作為一種重要的免疫效應(yīng)細胞,其在機體免疫中的作用不可忽視,巨噬細胞的正常凋亡對于其正常行使功能有著重要的意義。影響巨噬細胞凋亡的因素有很多,其中性激素是不可忽視的一個因素。我們以前的研究表明,睪酮能夠抑制感染了杜氏利什曼原蟲的小鼠骨髓巨噬細胞的凋亡,那么,睪酮直接作用于小鼠骨髓巨噬細胞時,對其調(diào)亡的影響如何呢?此外,已知Fas及FasL是介導(dǎo)細胞凋亡的一對膜蛋白,當(dāng)FasL與Fas結(jié)合時,可在數(shù)小時內(nèi)誘發(fā)Fas表達細胞凋亡,且巨噬細胞上有Fas的表達;并已知NO是存在于體內(nèi)有多種生物學(xué)活性的無機分子,它可以通過影響細胞物質(zhì)代謝和能量代謝而造成細胞死亡,且有報道認(rèn)為NO是誘導(dǎo)巨噬細胞凋亡的因子之一,故本研究的目的是觀察在睪酮的直接作用下,小鼠骨髓巨噬細胞會否發(fā)生凋亡;如果發(fā)生凋亡,那么在這個過程中,F(xiàn)as系統(tǒng)和NO是否參與了作用。 實驗方法: 1.動物M乏一5 ter Work e57BI丫6j小鼠購自,},...
