【摘要】：目的篩選K48聚泛素鏈在腫瘤壞死因子受體相關因子2(tumor necrosis factor receptor-associated facto r 2,T RA F2)上的主要修飾位點。方法比較不同物種間的T RA F2氨基酸序列,選出人T RA F2上保守率在90%以上的賴氨酸。構建人野生型TRAF2的表達載體及保守賴氨酸突變?yōu)榫彼岬耐蛔儽磉_載體。將不同的TRAF2表達載體與NF-κB熒光素酶報告基因表達載體共轉至293FT細胞中,通過熒光素酶檢測NF-κB激活情況。結果人TRAF2上共有8個保守率在90%以上的賴氨酸,酶切及測序結果證明本研究成功構建TRAF2野生型和突變型表達載體。NF-κB熒光素酶報告基因檢測證明TRAF2-K320可能是K48聚泛素鏈的主要修飾位點。結論 TRAF2-K320位點對TRAF2介導的NF-κB激活具有負向調控作用。
[Abstract]:Objective to screen the main modification sites of K48 polyubiquitin chain on tumor necrosis factor receptor related factor 2 (tumor necrosis factor receptor-associated facto r 2 T RA F 2. Methods the amino acid sequences of T RA F 2 in different species were compared, and lysine with a conservative rate of more than 90% on human T RA F 2 was selected. The expression vector of human wild-type TRAF2 and the mutant expression vector of conserved lysine to arginine were constructed. Different TRAF2 expression vectors and NF- 魏 B luciferase reporter gene expression vectors were co-transfected into 293FT cells, and the activation of NF- 魏 B was detected by luciferase. Results there were 8 lysines with a conservative rate of more than 90% in human TRAF2. Restriction endonuclease digestion and sequencing showed that TRAF2 wild-type and mutant expression vector. NF- 魏 B luciferase reporter gene was successfully constructed in this study. The results showed that TRAF2-K320 may be the main modification site of K48 polyubiquitin chain. Conclusion TRAF2-K320 site plays a negative role in the activation of NF- 魏 B mediated by TRAF2.
