B淋巴細(xì)胞刺激因子治療性疫苗的研究

【摘要】:B淋巴細(xì)胞刺激因子(B Lymphocyte Stimulator,BLyS)是1999年由瑞士和美國科學(xué)家發(fā)現(xiàn)的腫瘤壞死因子(Tumor Necrosis Factor,TNF)家族的一個新成員。該分子含有285個氨基酸,屬于Ⅱ型跨膜蛋白,編碼區(qū)位于染色體13q32-34。BLyS主要由T細(xì)胞和樹突狀細(xì)胞等髓系來源的細(xì)胞分泌。BLyS以膜結(jié)合以及可溶性兩種方式存在于機(jī)體中,兩者均具有生物學(xué)活性。 迄今為止,已經(jīng)發(fā)現(xiàn)了三種受體可以與BLyS結(jié)合,分別為BCMA、TACI和BAFF-R,由于缺少信號轉(zhuǎn)導(dǎo)序列,它們被劃歸為Ⅲ型膜蛋白。其中前兩者也可以與其他的分子結(jié)合,而BAFF-R則為BLyS特異性的受體,這些受體僅位于B細(xì)胞膜表面,使得BLyS的生物學(xué)活性——刺激B淋巴細(xì)胞的成熟以及增殖,有了高度的選擇性。 研究表明,BLyS在B細(xì)胞的存活和功能調(diào)節(jié)方面發(fā)揮著重要作用,它可以延長B細(xì)胞存活時間、促進(jìn)其增殖,從而導(dǎo)致B細(xì)胞增生和自身免疫狼瘡樣變化的出現(xiàn)。BLyS的過度表達(dá)是發(fā)生自身免疫性疾病,尤其是系統(tǒng)性紅斑狼瘡的重要因素之一。但是,對于BLyS的作用機(jī)制還不明了。

The molecule contains 285 amino acids and belongs to type II transmembrane protein. The coding region is located in the chromosome 13q32-34.BLyS secreted mainly by myeloid cells such as T cells and dendritic cells. BLyS exists in the organism in two ways: membrane binding and soluble. Both have biological activity.
