兔中性粒細(xì)胞防御素對(duì)A549細(xì)胞IL-8基因表達(dá)的影響

【摘要】:【背景】脊椎動(dòng)物防御素(defensins)是小分子的陽(yáng)離子抗菌多肽,它們廣泛分布于上皮細(xì)胞和吞噬細(xì)胞中。人和兔中性粒細(xì)胞的總蛋白中5%以上為防御素,它們除了具有直接的抗菌功能外還能作為免疫活性分子作用于上皮細(xì)胞和免疫細(xì)胞,參與天然免疫和獲得性免疫的調(diào)節(jié)。另外它們還可作為信號(hào)分子調(diào)節(jié)細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)過(guò)程,,促進(jìn)細(xì)胞的增殖、分化等。探索防御素的非抗菌功能是目前防御素研究的重點(diǎn)方向之一,本研究從mRNA和蛋白水平初步探討了兔中性粒細(xì)胞防御素(rabbits neutrophils antimicrobial peptides,NPs)對(duì)人Ⅱ型肺泡上皮細(xì)胞A549 IL-8表達(dá)的影響及其作用機(jī)制。 【方法與結(jié)果】向家兔腹腔注射1%酪蛋白350ml,構(gòu)建兔腹腔無(wú)菌性炎癥動(dòng)物模型。收集腹腔炎性滲出液,離心后棄上清,用雙蒸水破紅細(xì)胞,離心后超聲破碎白細(xì)胞,再次離心收集上清。上清經(jīng)Bio-Gel P-10低壓色譜凝膠層析柱分離純化后,用RP-HPLC法經(jīng)C18柱進(jìn)一步純化,
