【摘要】：目的探討一種改進的痛風性關節(jié)炎大鼠模型制備方法。方法將60只雄性SD大鼠平均分為兩個試驗進行研究。實驗一,將30只大鼠隨機分為傳統(tǒng)造模方法組和改進造模方法組,每組15只。其中傳統(tǒng)造模方法組于右踝關節(jié)前側(cè)穿刺注射生理鹽水50μL;改進造模方法組于大鼠右踝關節(jié)后側(cè)沿跟腱內(nèi)側(cè)以30°~40°方向插入至關節(jié)腔,注入生理鹽水50μL。各組大鼠于關節(jié)腔注射后0 h、12 h、24 h各取5只行步態(tài)評分和關節(jié)組織形態(tài)學檢查。實驗二,將30只大鼠隨機分為150μL組、100μL組和50μL組,每組10只,于大鼠右踝關節(jié)后側(cè)穿刺,分別將150μL、100μL和50μL歐乃派克注入關節(jié)腔并進行X線檢查。結(jié)果大鼠踝關節(jié)后側(cè)關節(jié)腔容積較大,便于進針;采用改進造模方法組的大鼠步態(tài)評分在12 h和24 h均較傳統(tǒng)造模方法組低,差異具有統(tǒng)計學意義(P0.05);改進的造模方法可避免對關節(jié)軟骨面的損傷,可防止因穿刺方法帶來的創(chuàng)傷性的關節(jié)炎性改變;不高于100μL的注藥劑量可使藥物存留在關節(jié)腔內(nèi)而不至于溢出。結(jié)論改進的痛風性關節(jié)炎大鼠模型制備方法較傳統(tǒng)方法對大鼠踝關節(jié)破壞小,有利于減少傳統(tǒng)方法對模型穩(wěn)定性帶來的干擾。
[Abstract]:Objective to explore an improved method for the preparation of gouty arthritis rat model. Methods 60 male SD rats were divided into two experiments on average. In experiment 1, 30 rats were randomly divided into traditional modeling group and improved model making group with 15 rats in each group. The traditional model group was injected with normal saline 50 渭 L at the anterior side of the right ankle joint, and the modified model group was inserted into the articular cavity at the posterior side of the right ankle joint at the direction of 30 擄(40 擄) along the medial side of the Achilles tendon and injected with normal saline (50 渭 L). The gait score and histomorphology of joints were examined in 5 rats in each group at 0 h and 12 h and 24 h after articular injection. In experiment 2, 30 rats were randomly divided into 150 渭 L group (100 渭 L) and 50 渭 L group (10 rats in each group). Results the volume of posterior articular cavity of the ankle joint in rats was larger, and the gait score of the modified model group was lower than that of the traditional model group at 12 h and 24 h, and the gait score of the modified model group was lower than that of the traditional model group. The difference was statistically significant (P0.05); the improved modeling method could avoid the injury of articular cartilage and prevent the traumatic arthritis changes caused by puncture method. A dose of no more than 100 渭 L allows the drug to remain in the articular cavity without overflowing. Conclusion the modified gouty arthritis rat model has less damage to the ankle joint than the traditional method, and it is helpful to reduce the interference of the traditional method to the stability of the model.
