【摘要】: 狂犬病病毒屬?gòu)棤畈《究频目袢《緦?它屬于單鏈RNA病毒。是引起人畜共患性傳染病-狂犬病的病原體。自70年代以來(lái),有關(guān)狂犬病街毒株及固定毒株在動(dòng)物體內(nèi)及培養(yǎng)細(xì)胞內(nèi)的病毒抗原的熒光抗體染色及其病毒超微結(jié)構(gòu)的研究陸續(xù)有報(bào)道[1-3]。但廣泛應(yīng)用于生產(chǎn)狂犬細(xì)胞培養(yǎng)疫苗的固定毒aG株在原代地鼠腎細(xì)胞內(nèi)病毒復(fù)制部位、病毒增殖動(dòng)態(tài)、病毒的超微結(jié)構(gòu)、病毒形態(tài)發(fā)生以及光鏡下病毒抗原定位與電鏡下病毒復(fù)制部位的關(guān)系等報(bào)道很少。掌握病毒感染細(xì)胞后的生物學(xué)過(guò)程對(duì)狂犬病的預(yù)防、診斷、治療都有重要意義。另外這方面的基礎(chǔ)理論資料對(duì)改進(jìn)生產(chǎn)工藝,提高狂犬疫苗產(chǎn)量和質(zhì)量也有指導(dǎo)意義,同時(shí)也為生產(chǎn)和檢定部門提供敏感性高、特異性強(qiáng),并且簡(jiǎn)單快速的狂犬病毒抗原檢測(cè)方法。為此,本論文利用現(xiàn)代免疫酶組織化學(xué)及電鏡技術(shù)進(jìn)行了系統(tǒng)試驗(yàn)。本實(shí)驗(yàn)著重對(duì)免疫組織化學(xué)技術(shù)中常用的免疫熒光法和免疫酶組織化學(xué)法(ABC法)進(jìn)行對(duì)比試驗(yàn)。并對(duì)光鏡和電鏡觀察結(jié)果進(jìn)行了比較。
[Abstract]:Rabies virus of the family Granuloviridae, belonging to a single-stranded RNA virus. Is the cause of zoonotic sexually transmitted diseases-rabies pathogens. Since the 1970s, the studies on fluorescent antibody staining and ultrastructure of rabies street virus strains and stationary strains in animal and cultured cells have been reported one after another. But it is widely used in producing rabies cell culture vaccine. The viral replication site, virus proliferation dynamics and virus ultrastructure of the virus in primary hamster kidney cells are the fixed virus AG strain, which is widely used in the production of rabies cell culture vaccine. There are few reports on the relationship between viral morphogenesis and the localization of virus antigen under light microscope and the replication site of virus under electron microscope. It is important to master the biological process of virus infection for the prevention, diagnosis and treatment of rabies. In addition, the basic theoretical data in this respect also have guiding significance for improving the production process and improving the production and quality of rabies vaccine, and also provide high sensitivity and specificity for the production and verification departments. And simple and rapid detection method of rabies virus antigen. Therefore, a systematic experiment was carried out by using modern immunoenzyme histochemistry and electron microscopy. In this experiment, the immunofluorescence method and the immuno-enzyme histochemical method (ABC method), which are commonly used in immunohistochemistry, were studied. The results of light microscope and electron microscope were compared.
