胎盤源間充質(zhì)干細(xì)胞對T淋巴細(xì)胞體外增殖和細(xì)胞因子分泌的影響

【摘要】: 目的:間充質(zhì)干細(xì)胞(mesenchymal stem cells,MSCs)具有自我更新和多向分化的潛能,被認(rèn)為在移植治療、基因治療、細(xì)胞治療、組織工程等領(lǐng)域具有廣闊的臨床應(yīng)用前景。而MSCs的來源以及MSCs的免疫調(diào)節(jié)機制是亟需解決的兩大問題。本實驗的目的旨在從人胎盤組織中分離培養(yǎng)獲得間充質(zhì)干細(xì)胞,并進一步研究其對T淋巴細(xì)胞體外增殖和細(xì)胞因子分泌的影響。 方法:首先從胎盤組織中分離培養(yǎng)胎盤間充質(zhì)干細(xì)胞(PMSCs),在體外觀測其形態(tài),并通過細(xì)胞表面抗原表達(dá)、分化潛能等特征進行鑒定;然后將體外分離培養(yǎng)擴增的PMSCs,按照不同比例加入雙向混合淋巴細(xì)胞培養(yǎng)體系(mixed lymphocyte reaction, MLR)中,共同培養(yǎng)3d后,用ELISA法檢測各組MLC上清中細(xì)胞因子IL-2、IL-4與IFN-γ的含量以及共同培養(yǎng)6d后,測定T淋巴細(xì)胞的增殖。 結(jié)果:1.從人胎盤組織中分離培養(yǎng)獲得間充質(zhì)干細(xì)胞,具有與骨髓間充質(zhì)干細(xì)胞(BMSCs)極為相似的細(xì)胞形態(tài)及細(xì)胞表面標(biāo)志,表達(dá)CD29、CD44和CD105,不表達(dá)CD34、CD45,、CD106和HLA-DR。并且還具有與BMSCs相似的跨胚層分化能力,能在一定條件下被誘導(dǎo)分化出神經(jīng)元樣細(xì)胞。2.PMSCs與同種異體混合淋巴細(xì)胞共同培養(yǎng)后,能抑制T淋巴細(xì)胞的體外增殖和影響細(xì)胞因子IL-2、IL-4與IFN-γ的分泌。 結(jié)論:胎盤組織是MSCs的有效來源,PMSCs具有與BMSCs相似的生物學(xué)特性及免疫調(diào)節(jié)機制。它為間充質(zhì)干細(xì)胞的研究提供了又一重要的細(xì)胞來源。
[Abstract]:Objective: mesenchymal stem cells (MSCs) have the potential of self-renewal and multi-differentiation. They are widely used in transplantation therapy, gene therapy, cell therapy, tissue engineering and so on. The origin of MSCs and the immunomodulation mechanism of MSCs are two problems that need to be solved. The aim of this study was to isolate and culture mesenchymal stem cells from human placenta and to investigate the effects of mesenchymal stem cells on T lymphocyte proliferation and cytokine secretion in vitro. Methods: placental mesenchymal stem cells (PMSCs) were isolated and cultured from placenta tissue. The morphology of PMSCs was observed in vitro. After 3 days of co-culture, the levels of cytokines IL-2, IL-4 and IFN- 緯 in the supernatant of MLC were detected by Elisa and co-cultured for 6 days. The proliferation of T lymphocytes was measured. The result is 1: 1. Mesenchymal stem cells (MSCs) isolated from human placental tissue have similar cell morphology and cell surface markers to bone marrow mesenchymal stem cells (BMSCs). They express CD29, CD44 and CD105, and do not express CD34, CD45, CD106 and HLA-DR. It also has the ability of transdermal differentiation similar to that of BMSCs, and can be induced to differentiate into neuron-like cells. 2. PMSCs can be co-cultured with allogenic mixed lymphocytes. It can inhibit the proliferation of T lymphocytes in vitro and affect the secretion of cytokines IL-4 and IFN- 緯. Conclusion: placental tissue is an effective source of MSCs, PMSCs have similar biological characteristics and immunomodulatory mechanism as BMSCs. It provides another important cell source for the study of mesenchymal stem cells.
