長(zhǎng)期飲用硝酸釤對(duì)小鼠免疫功能影響的實(shí)驗(yàn)研究

【摘要】:目的:研究稀土的長(zhǎng)期攝入對(duì)機(jī)體免疫功能是否存在影響,為制定稀土使用安全標(biāo)準(zhǔn)和評(píng)價(jià)稀土生物效應(yīng)提供實(shí)驗(yàn)依據(jù)。本實(shí)驗(yàn)以不同劑量(5、50、500、2000mg/L)硝酸釤染毒3個(gè)月的ICR小鼠為對(duì)象,通過(guò)染毒過(guò)程中體重的動(dòng)態(tài)變化、臟器指數(shù)、遲發(fā)型變態(tài)反應(yīng)、E花結(jié)形成率、免疫球蛋白含量及腹腔巨噬細(xì)胞吞噬功能、紅細(xì)胞受體花環(huán)研究了小鼠細(xì)胞免疫功能、體液免疫功能、巨噬細(xì)胞吞噬功能、紅細(xì)胞免疫吸附功能的變化;另外通過(guò)對(duì)血清Sm濃度及部分與免疫功能密切相關(guān)的金屬元素的濃度、血清抗氧化能力測(cè)定及紅細(xì)胞形態(tài)觀(guān)察初步探討可能存在的硝酸釤免疫毒性機(jī)制。 結(jié)果:1.一般體征檢查結(jié)果表明小鼠在染毒過(guò)程中各組間體重?zé)o明顯差異(P>0.05)。雄鼠500mg·L~(-1)和2000mg·L~(-1)劑量組脾指數(shù)明顯低于對(duì)照組(P<0.05),其它臟器指數(shù)與對(duì)照組相比無(wú)統(tǒng)計(jì)學(xué)意義,推測(cè)脾對(duì)硝酸釤作用更敏感。各劑量組免疫器官肉眼觀(guān)察與對(duì)照組無(wú)明顯區(qū)別;隨染毒劑量增高,脾組織結(jié)構(gòu)發(fā)生變化,超微結(jié)構(gòu)觀(guān)察發(fā)現(xiàn)淋巴細(xì)胞核形變、線(xiàn)粒體腫脹空化、核周間隙增寬、巨細(xì)胞增多甚至細(xì)胞解體。 2.雌鼠在50mg·L~(-1)、500mg·L~(-1)劑量組的足跖腫脹度明顯低于對(duì)照組(P<0.05);而雄鼠除5mg·L~(-1)劑量組外,各組腫脹度均顯著下降,,2000mg·L~(-1)劑量組腫脹度僅為對(duì)照組的60%。當(dāng)硝酸釤濃度升高至2000mg·L~(-1)時(shí),雌雄鼠的E花結(jié)形成率均被嚴(yán)重抑制(P<0.01),可見(jiàn)飲用硝酸釤損傷了機(jī)體建立特異性細(xì)胞免疫的能力。 3.雌鼠除5mg·L~(-1)組外,其余各組溶血空斑形成數(shù)顯著下降(P<0.01),而雄鼠即使在最低劑量組PFC也出現(xiàn)明顯變化(P<0.05)。在一定濃度范圍內(nèi)(5~500mg·L~(-1)),硝酸釤可能會(huì)抑制更多抗體(IgG)產(chǎn)生,隨著染毒劑量的增加,這種抑制作用減弱。硝酸釤的攝入對(duì)小鼠體液免疫應(yīng)
[Abstract]:Objective: To study the effect of the long-term intake of rare earth on the immune function of the body, to provide an experimental basis for the formulation of the safety standard for the use of rare earth and the evaluation of the biological effect of rare earth. In this experiment, the ICR mice infected with samarium nitrate at different doses (5,505002000mg / L) were used for 3 months, through the dynamic changes of body weight and the organ index. The late onset allergy, the formation rate of E flower knot, the content of immunoglobulin and the phagocytosis of peritoneal macrophages. The erythrocyte receptor rosette studied the cellular immune function, the humoral immune function, the macrophage phagocytosis and the changes of the red cell immune adsorption function, and the serum Sm concentration and part of the immune function were closely related to the serum concentration and part of the serum. The concentrations of metal elements, serum antioxidant capacity and morphology of erythrocytes were preliminarily explored to explore the possible mechanism of immune toxicity of samarium nitrate.
Results: 1. the results of general physical examination showed that there was no significant difference in weight between the mice during the process of exposure (P > 0.05). The spleen index of the male rats 500mg L~ (-1) and 2000mg. L~ (-1) dose group was significantly lower than that of the control group (P < 0.05). The other visceral index had no statistical significance compared with the control group, and the spleen was more sensitive to samarium nitrate. There was no obvious difference between the immune organs and the control group. With the increase of the dose, the structure of the spleen was changed. The ultrastructural observation revealed the deformation of the lymphocyte nucleus, the swelling and cavitation of mitochondria, the widening of the perinuclear space, the increase of giant cells and even the disintegration of the cells.
The feet swelling degree of 2. female rats in 50mg. L~ (-1) and 500mg. L~ (-1) dose group was significantly lower than that of the control group (P < 0.05), but the swelling degree of the male rats was significantly lower than the 5mg. L~ (-1) dose group. The swelling degree of the 2000mg L~ dose group was only the control group when the concentration of samarium nitrate increased to the conclusion. Severely inhibited (P < 0.01), it is obvious that drinking samarium nitrate damages the body's ability to establish specific cellular immunity.
3. female rats in addition to 5mg L~ (-1) group, the formation of hemolytic plaque in the rest of the other groups decreased (P < 0.01), and the male rats had obvious changes (P < 0.05) even in the lowest dose group (P < 0.05). In a certain concentration range (5 to 500mg. L~ (-1)), samarium nitrate could inhibit more antibodies (IgG), and this inhibition weakened with the increase of the dose.
