松果體及褪黑素在大鼠胸腺T細(xì)胞分化發(fā)育中的作用

【摘要】:自從1926年Berman提出松果體與胸腺相關(guān)的概念以來(lái),許多實(shí)驗(yàn)研究表明松果體與胸腺之間存在較明顯的功能聯(lián)系,然而迄今為止的研究絕大多數(shù)為體外實(shí)驗(yàn),且研究手段較局限,因而松果體與胸腺結(jié)構(gòu)和功能的關(guān)系及其相關(guān)機(jī)制仍然不明確。本研究運(yùn)用大鼠松果體摘除動(dòng)物模型,從以下四個(gè)方面探討松果體及其褪黑素對(duì)大鼠胸腺T細(xì)胞分化發(fā)育的作用及其可能的機(jī)制。 一、松果體及褪黑素對(duì)大鼠胸腺T細(xì)胞分化發(fā)育的影響 有關(guān)松果體及其褪黑素對(duì)胸腺T細(xì)胞分化發(fā)育過(guò)程中不同亞群的影響及其可能的機(jī)制尚未見(jiàn)報(bào)道。本部分運(yùn)用免疫細(xì)胞化學(xué)、計(jì)算機(jī)圖像分析以及CD_4、CD_8雙色直標(biāo)單抗反應(yīng)法流式細(xì)胞術(shù)檢測(cè)松果體摘除及補(bǔ)充褪黑素后胸腺和血液T細(xì)胞亞群的比例,觀察松果體摘除后胸腺和血液T細(xì)胞CD4/8不同亞群的變化,以及補(bǔ)充褪黑素的影響,探討松果體及其褪黑素在大鼠胸腺T細(xì)胞分化發(fā)育中的作用。 結(jié)果顯示松果體摘除后4周時(shí)大鼠體重下降明顯(P0.05),術(shù)后8周胸腺重量減輕顯著。褪黑素補(bǔ)充能在一定程度上緩解松果體摘除引起的體重和胸腺重量下降趨勢(shì),高劑量組尤為明顯。松果體摘除后8周胸腺皮質(zhì)CD4~+陽(yáng)性細(xì)胞面積減少具統(tǒng)計(jì)學(xué)意義,補(bǔ)充褪黑素后呈劑量依賴(lài)性恢復(fù)。髓質(zhì)CD8~+細(xì)胞面積術(shù)后4周明顯升高,補(bǔ)充褪黑素后略有下降,但其升高仍然具統(tǒng)計(jì)學(xué)意義。流式細(xì)胞術(shù)檢測(cè)結(jié)果顯示松果體摘除后4周胸腺內(nèi)CD4~+細(xì)胞比例以及雙陽(yáng)性、雙陰性細(xì)胞比例變化不明顯,而CD8~+細(xì)胞比例則下降明顯,至術(shù)后8周CD8~+細(xì)胞比例則上升,雙陰性細(xì)胞比例降低。補(bǔ)充褪黑素4周時(shí)CD8~+細(xì)胞比例呈劑量依賴(lài)性升高,高劑量組無(wú)論4周還是8周均有顯著性意義,雙陽(yáng)性細(xì)胞比例則降低。松
[Abstract]:Since Berman put forward the concept of pineal gland and thymus in 1926, many experimental studies have shown that there is an obvious functional relationship between pineal gland and thymus. Therefore, the relationship between pineal gland and thymus structure and function and its related mechanism are still unclear. In this study, the effects of pineal gland and melatonin on the differentiation and development of T cells in rat thymus were studied from the following four aspects using the rat model of pinealectomy. First, Effects of pineal gland and melatonin on the differentiation and development of T cells in rat thymus the different subtypes of pineal gland and melatonin on the differentiation and development of thymus T cells The influence of the group and its possible mechanism have not been reported. In this part, immunocytochemistry, computer image analysis and flow cytometry were used to detect T lymphocyte subsets in thymus and blood after pinealectomy and melatonin supplementation. To observe the changes of CD _ 4 / 8 subsets of T cells in thymus and blood after pinealectomy and the effects of melatonin supplementation on the differentiation and development of T cells in rat thymus. The results showed that the weight of the thymus decreased significantly at 4 weeks after pinealectomy (P0.05), and the weight of thymus decreased significantly at 8 weeks after operation. The decrease of weight and thymus weight caused by pinealectomy was alleviated to some extent by melatonin supplementation, especially in the high dose group. The area of CD4- positive cells in thymic cortex decreased significantly 8 weeks after pinealectomy and recovered in a dose-dependent manner after supplementation of melatonin. The area of CD8 ~ cells in medulla increased significantly 4 weeks after operation, and decreased slightly after melatonin supplementation, but the increase was still statistically significant. The results of flow cytometry showed that the percentage of CD4 ~ cells and double positive cells in thymus of pineal gland 4 weeks after pinealectomy did not change significantly, but the proportion of CD8 ~ cells decreased significantly, but the proportion of CD8 ~ cells increased at 8 weeks after operation. The proportion of double negative cells decreased. The percentage of CD8- cells increased in a dose-dependent manner at the 4th week after melatonin supplementation.
