表達(dá)hCGβ-C3d3融合蛋白的乳酸桿菌活疫苗粘膜免疫增強抗hCGβ免疫效應(yīng)機制

【摘要】:目的:將含有融合基因hCGβ-C3d3的質(zhì)粒轉(zhuǎn)化干酪乳酸桿菌,對比表達(dá)hCGβ的重組乳酸桿菌活疫苗Lb.hCGβ或Lb.hCGβ-C3d3經(jīng)不同粘膜途徑接種不同品系小鼠,,產(chǎn)生抗hCGβ免疫應(yīng)答,驗證C3d3在表達(dá)hCGβ重組乳酸桿菌活疫苗粘膜免疫中的佐劑作用;探討C3d3在表達(dá)hCGβ重組乳酸桿菌活疫苗粘膜免疫中增強hCGβ免疫原性的效應(yīng)機制。 方法:用分子生物學(xué)技術(shù)構(gòu)建質(zhì)粒pIlac.hCGβ-C3d3,電穿孔轉(zhuǎn)化干酪乳酸桿菌(Lactobacillus casei)CECT 5276,挑選轉(zhuǎn)化后的耐藥菌落,在含紅霉素的MRS培養(yǎng)基中培養(yǎng),0.5%乳糖誘導(dǎo)后,在不同時間取上清,化學(xué)發(fā)光法檢測hCGβ,Western blot鑒定表達(dá)產(chǎn)物,細(xì)胞免疫化學(xué)鑒定融合蛋白。用10~8,10~9,10~(10)個重組乳酸桿菌Lb.hCGβ或Lb.hCGβ-C3d3分別經(jīng)消化道、呼吸道和陰道粘膜接種6-8周齡雌性BALB/c和C57BL/6小鼠,3周后用相同劑量加強免疫一次。間接ELISA檢測初次接種后2—8周血清和陰道灌洗液中抗hCGβ IgG和IgA抗體滴度;以及抗血清中和hCG抗原的活性。用10~(10)個重組乳酸桿菌Lb.hCGβ或Lb.hCGβ-C3d3分別經(jīng)口腔、鼻腔和陰道粘膜接種6-8周齡雌性BALB/c和C57BL/6小鼠,分離免疫小鼠脾、子[⒁醯樂械牧馨拖赴,留|較赴欠治銎銪和T細(xì)胞的增殖情況,ELISpot分析其分泌抗hCGβ IgG和IgA抗體的抗體分泌細(xì)胞克隆數(shù)。 結(jié)果:經(jīng)測序質(zhì)粒構(gòu)建成功;轉(zhuǎn)化成功的重組干酪乳酸桿菌在乳糖誘導(dǎo)下可分泌融合蛋白hCGβ-C3d3。在不同品系的小鼠體內(nèi)10~9,10~(10) Lb.hCGβ可誘導(dǎo)相似的免疫應(yīng)答,而10~8誘導(dǎo)的抗體滴度較低。三種粘膜免疫途徑誘導(dǎo)的免疫應(yīng)答相似。加強免疫后10~9和10~(10) Lb.hCGβ誘導(dǎo)的抗血清中和hCG抗原的能力>100ng/ml。陰道粘膜接種誘導(dǎo)的最高抗體滴度不低于、甚至優(yōu)于其他兩種粘膜接種途徑。用Lb.hCGβ-C3d3接種誘導(dǎo)的最高效價比Lb.hCGβ高2—100倍,加強接種后抗血清能有效中和hCG抗原的生物學(xué)活性。重組乳酸桿菌至少可在BALB/c小鼠陰道粘膜內(nèi)存活3周。與Lb.hCGβ相比,Lb.hCGβ-C3d3粘膜接種后T和B細(xì)胞增殖能力明顯增強(p<0.05)。脾臟內(nèi)IgG及IgA抗體分泌細(xì)胞個數(shù)在Lb.hCGβ或Lb.hCGβ-C3d3各種免疫途徑間無顯著差異(p>0.05);但Lb.hCGβ-C3d3粘膜接種后子[耙醯濫贗gG及IgA抗體分泌細(xì)胞個數(shù)均顯著高于Lb.hCGβ(p<0.05)。 結(jié)論:實現(xiàn)了重組乳酸桿菌Lb.hcGβ-C3d3分泌性表達(dá)融合蛋白hCGβ-C3d3,培養(yǎng)4小時后0.5%乳糖誘導(dǎo)是重組菌表達(dá)hCGβ的優(yōu)化條件。重組乳酸桿菌活疫苗Lb.hCGβ可經(jīng)粘膜免疫誘導(dǎo)有效的免疫應(yīng)答,產(chǎn)生的應(yīng)答能力與免疫劑量有關(guān)。陰道粘膜接種可能是一種誘導(dǎo)生殖道粘膜免疫的較好途徑。用表達(dá)hCGβ重組乳酸桿菌活疫苗粘膜免疫不同品系小鼠,C3d3均能增強機體對hCGβ抗原的體液免疫應(yīng)答。顯著刺激 T和B細(xì)胞增殖;顯著增加子宮及陰道內(nèi)抗hCGβ IgG和IgA抗體的抗體分泌細(xì)胞克隆數(shù),可能是C3d3在粘膜免疫中增強體液免疫的機制。
[Abstract]:Objective : To transform the recombinant lactobacillus live vaccine ( Lb . hCG尾 or Lb . hCG尾 - C3d3 ) containing the fusion gene hCG尾 - C3d3 into different strain mice by different mucosal routes to produce anti - hCG尾 immune response , and to verify the adjuvant role of C3d3 in the expression of hCG尾 recombinant lactobacillus live vaccine mucosal immunity ;
The effect mechanism of C3d3 on enhancing the immunogenicity of hCG尾 in the mucosal immunity of hCG尾 recombinant Lactobacillus live vaccine was investigated .
Methods : The plasmid pIlac . hCG尾 - C3d3 was constructed by molecular biology technique . The transformed drug resistant colonies were selected by electroporation .
The anti - hCG尾 or Lb . hCG尾 - C3d3 were inoculated 6 - 8 weeks old female BALB / c and C57BL / 6 mice respectively via oral cavity , nasal cavity and vaginal mucosa .
Results : The sequencing plasmid was successfully constructed .
There was no significant difference in the number of IgG and IgA antibody - secreting cells in BALB / c mice than that of Lb . hCG尾 - C3d3 , and the number of IgG and IgA antibody - secreting cells in spleen was not significantly different from that of Lb . hCG尾 or Lb . hCG尾 - C3d3 ( p > 0.05 ) .
But Lb . hCG尾 - C3d3 mucosal vaccination was significantly higher than that of Lb . hCG尾 ( p < 0.05 ) .
Conclusion : The recombinant lactobacillus Lb . hcG . beta . - C3d3 secretory expression fusion protein hCG尾 - C3d3 is expressed . The optimal conditions for the expression of hCG尾 in recombinant bacteria after 4 hours of culture . The recombinant Lactobacillus live vaccine Lb . hCG尾 can induce an effective immune response via mucosal immunity .
T and B cell proliferation ;
It is possible to increase the number of antibody secreting cells in uterus and intravaginal anti - hCG beta IgG and IgA antibody , which may be the mechanism of C3d3 to enhance humoral immunity in mucosal immunity .
