日本血吸蟲復(fù)合B細(xì)胞表位抗原的研究

【摘要】: 血吸蟲病是一種嚴(yán)重危害人類健康的人畜共患寄生蟲病。我國(guó)是全球血吸蟲病危害最嚴(yán)重的國(guó)家(埃及、蘇丹、中國(guó)、巴西)之一,我國(guó)流行日本血吸蟲病,主要流行于長(zhǎng)江流域及以南的12個(gè)省(市、自治區(qū))的434個(gè)縣(市、區(qū)),受威脅的人口約2億。經(jīng)50多年防治血吸蟲病的不懈努力,我國(guó)血吸蟲病防治工作已取得了巨大的成績(jī)。但目前全國(guó)仍有109個(gè)縣(市、區(qū))有血吸蟲病流行,病人數(shù)達(dá)84.25萬,病畜約2.41萬頭。 血吸蟲病的診斷在血吸蟲病防治中起著極為重要的作用。隨著血吸蟲病防治措施的實(shí)施,流行區(qū)的感染度和感染率明顯下降,以至血吸蟲病經(jīng)典的寄生蟲學(xué)診斷方法-糞檢的敏感性明顯下降,并且由于糞便檢查費(fèi)工、費(fèi)時(shí)、費(fèi)力,疫區(qū)群眾的依從性逐年下降,這種經(jīng)典的寄生蟲學(xué)診斷技術(shù)在現(xiàn)場(chǎng)大規(guī)模查病中的應(yīng)用受到很大的限制。由于免疫診斷技術(shù)具有操作簡(jiǎn)便、敏感性較高、特異性較好等優(yōu)點(diǎn),因此發(fā)展免疫學(xué)診斷方法來代替或補(bǔ)充糞檢為廣大專業(yè)人員所采納。常用的免疫學(xué)診斷方法所采用的檢測(cè)抗原多為血吸蟲成蟲或蟲卵抗原,這些抗原成分復(fù)雜且成本較高;而用基因工程方法制備完整的抗原雖可以獲得較純的抗原,但常常由于表達(dá)效率不高或純化困難,因而限
[Abstract]:Schistosomiasis is a zoonotic parasitic disease that seriously endangers human health. China is one of the most serious countries (Egypt, Sudan, China, Brazil) in the world. Schistosomiasis japonicum is prevalent in China, mainly in 434 counties (cities) in 12 provinces (autonomous regions) south of the Yangtze River valley. The population at risk is about 200 million. After more than 50 years of unremitting efforts to control schistosomiasis, great achievements have been made in the prevention and control of schistosomiasis in China. But at present, there are 109 counties (cities and districts) with schistosomiasis epidemic in China, the number of people is 842500 and the number of animals is about 24100. The diagnosis of schistosomiasis plays an important role in the prevention and control of schistosomiasis. With the implementation of schistosomiasis control measures, the infection degree and infection rate in endemic areas decreased obviously, and the sensitivity of fecal examination, the classic parasitological diagnostic method of schistosomiasis, decreased obviously, and it was time-consuming and laborious because of the cost of stool examination. The compliance of the masses in epidemic areas is declining year by year. The application of this classical parasitological diagnosis technique in the field of large-scale disease detection is greatly limited. Because of the advantages of simple operation, high sensitivity, good specificity and so on, immunological diagnostic methods have been developed to replace or supplement fecal examination. Most of the commonly used immunological diagnostic antigens are the adult or egg antigens of Schistosoma japonicum, which are complicated and costly, but the relatively pure antigens can be obtained by using genetic engineering methods to prepare complete antigens. However, due to the low efficiency of expression or the difficulty of purification, it is often limited.
