【摘要】:PTEN基因(phosphatase and tension homology deletion on chromosome 10, PTEN)定位在10q23,編碼一種具有磷酸酶活性的蛋白,此蛋白與酪氨酸磷酸酶與細(xì)胞骨架蛋白tensin和auxilin高度同源。PTEN是一種抑癌基因,PTEN體細(xì)胞突變可出現(xiàn)在許多惡性腫瘤的早期和晚期,如前列腺癌、腦膠質(zhì)瘤、肝癌、腎癌、子宮內(nèi)膜癌的發(fā)生、進(jìn)展。除了抑癌作用外,PTEN還參與細(xì)胞的凋亡、增殖、遷徙和PI3K—AKT信號通路。但近來有研究表明,PTEN與細(xì)胞的體積大小有密切關(guān)系,尤其是神經(jīng)元和心肌細(xì)胞肥大。文獻(xiàn)報道,PTEN的過表達(dá)和失活同樣都會影響果蠅的眼睛和翅膀體積,敲除PTEN的小鼠出現(xiàn)大頭畸形,而在人類大頭畸形的患者中檢測到PTEN的缺失。影響細(xì)胞體積大小的關(guān)鍵因素是細(xì)胞內(nèi)DNA含量;其次,是細(xì)胞所處的外界環(huán)境;另外一個影響因素是溫度。雖然人們一直對細(xì)胞體積大小很感興趣,但其內(nèi)在的基因調(diào)控系統(tǒng)至今仍未闡明。 在人體的正常組織和腫瘤組織中都有許多不同類型的大細(xì)胞,如人骨巨細(xì)胞瘤中(giant cell tumor of bone,GCT)的破骨樣多核巨細(xì)胞、卵巢中的
[Abstract]:The PTEN gene phosphatase and tension homology deletion on chromosome 10 (PTEN), located at 10q23, encodes a protein with phosphatase activity. This protein is highly homologous to tyrosine phosphatase and cytoskeletal proteins tensin and auxilin. PTEN somatic mutations may occur in the early and late stages of many malignant tumors, such as prostate cancer, glioma, liver cancer, renal cell carcinoma. The occurrence and progression of endometrial carcinoma. PTEN is involved in cell apoptosis, proliferation, migration and PI3K-AKT signaling. However, recent studies have shown that PTEN is closely related to the size of the cells, especially the hypertrophy of neurons and cardiomyocytes. It has been reported that both overexpression and inactivation of PTEN can affect the size of the eyes and wings of Drosophila melanogaster, and the large head malformation is found in the mice knocking out PTEN, while the deletion of PTEN is detected in the patients with human macrocephaly. The key factor affecting the cell volume is the content of DNA in the cell; secondly, the external environment of the cell; and the other factor is the temperature. Although people have always been interested in the size of the cell, its inherent gene regulation system has not been clarified. There are many different types of large cells in normal and tumor tissues of the human body, such as osteoclast-like multinucleated giant cells in giant cell tumor of human bone, osteoclastic multinucleated giant cells in ovary.
