抑制消減雜交技術(shù)篩選骨髓間充質(zhì)干細胞誘導分化前后差異表達基因

【摘要】: 骨髓間充質(zhì)干細胞(BMSCs)具有高度可塑性,在一定誘導條件下具有向成骨細胞、成軟骨細胞、肌腱細胞、脂肪細胞以及基質(zhì)細胞等中胚層細胞分化的能力,最近研究表明中胚層來源的BMSCs還可向外胚層來源的神經(jīng)元分化。 本研究在應用流式細胞術(shù)成功分選出大鼠骨髓間充質(zhì)干細胞的基礎(chǔ)上,采用視網(wǎng)膜細胞/骨髓間充質(zhì)干細胞共培養(yǎng)系統(tǒng),模擬視網(wǎng)膜體內(nèi)發(fā)育的微環(huán)境,誘導骨髓間充質(zhì)干細胞分化為視網(wǎng)膜神經(jīng)節(jié)樣細胞。經(jīng)免疫組化、神經(jīng)細胞特異性抗體及流式細胞儀檢測,鑒定細胞分化水平;再通過抑制性消減雜交(SSH)技術(shù)構(gòu)建骨髓間充質(zhì)干細誘導前后的cDNA消減文庫,篩選BMSCs向視網(wǎng)膜神經(jīng)節(jié)樣細胞分化過程中的差異表達基因。結(jié)果顯示,利用SSH技術(shù)篩選出骨髓間充質(zhì)干細胞在視網(wǎng)膜細胞誘導前后的差異表達基因GAP43,RGS等,上述研究結(jié)果表明BMSCs在誘導后表達某些視網(wǎng)膜神經(jīng)節(jié)細胞所特有的基因,從而為轉(zhuǎn)基因方法誘導BMSCs向視網(wǎng)膜神經(jīng)節(jié)細胞定向分化及其功能性研究奠定實驗基礎(chǔ)。
[Abstract]:Bone marrow mesenchymal stem cells (BMSCs) have the ability to differentiate into mesodermal cells such as osteoblasts, chondroblasts, tendon cells, adipocytes and stromal cells. Recent studies have shown that BMSCs derived from mesoderm can also differentiate into neurons derived from ectoderm. On the basis of the successful selection of rat bone marrow mesenchymal stem cells by flow cytometry, a retinal / bone marrow mesenchymal stem cell co-culture system was used to simulate the microenvironment of retinal development in vivo. Bone marrow mesenchymal stem cells were induced to differentiate into retinal ganglion like cells. The differentiation level of the cells was identified by immunohistochemistry, neuronal specific antibody and flow cytometry, and the cDNA subtractive library was constructed by suppression subtractive hybridization before and after fine induction of bone marrow mesenchymal stem. To screen the differentially expressed genes during the differentiation of BMSCs into retinal ganglion like cells. The results showed that the differentially expressed genes GAP43 and RGS of bone marrow mesenchymal stem cells (BMSCs) were screened by SSH technique before and after induction. The results showed that BMSCs expressed some specific genes in retinal ganglion cells after induction. The results provide experimental basis for the study of the differentiation and function of BMSCs into retinal ganglion cells by transgenic method.
