抑制消減雜交技術(shù)篩選骨髓間充質(zhì)干細(xì)胞誘導(dǎo)分化前后差異表達(dá)基因

【摘要】: 骨髓間充質(zhì)干細(xì)胞(BMSCs)具有高度可塑性,在一定誘導(dǎo)條件下具有向成骨細(xì)胞、成軟骨細(xì)胞、肌腱細(xì)胞、脂肪細(xì)胞以及基質(zhì)細(xì)胞等中胚層細(xì)胞分化的能力,最近研究表明中胚層來(lái)源的BMSCs還可向外胚層來(lái)源的神經(jīng)元分化。 本研究在應(yīng)用流式細(xì)胞術(shù)成功分選出大鼠骨髓間充質(zhì)干細(xì)胞的基礎(chǔ)上,采用視網(wǎng)膜細(xì)胞/骨髓間充質(zhì)干細(xì)胞共培養(yǎng)系統(tǒng),模擬視網(wǎng)膜體內(nèi)發(fā)育的微環(huán)境,誘導(dǎo)骨髓間充質(zhì)干細(xì)胞分化為視網(wǎng)膜神經(jīng)節(jié)樣細(xì)胞。經(jīng)免疫組化、神經(jīng)細(xì)胞特異性抗體及流式細(xì)胞儀檢測(cè),鑒定細(xì)胞分化水平;再通過(guò)抑制性消減雜交(SSH)技術(shù)構(gòu)建骨髓間充質(zhì)干細(xì)誘導(dǎo)前后的cDNA消減文庫(kù),篩選BMSCs向視網(wǎng)膜神經(jīng)節(jié)樣細(xì)胞分化過(guò)程中的差異表達(dá)基因。結(jié)果顯示,利用SSH技術(shù)篩選出骨髓間充質(zhì)干細(xì)胞在視網(wǎng)膜細(xì)胞誘導(dǎo)前后的差異表達(dá)基因GAP43,RGS等,上述研究結(jié)果表明BMSCs在誘導(dǎo)后表達(dá)某些視網(wǎng)膜神經(jīng)節(jié)細(xì)胞所特有的基因,從而為轉(zhuǎn)基因方法誘導(dǎo)BMSCs向視網(wǎng)膜神經(jīng)節(jié)細(xì)胞定向分化及其功能性研究奠定實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Bone marrow mesenchymal stem cells (BMSCs) have the ability to differentiate into mesodermal cells such as osteoblasts, chondroblasts, tendon cells, adipocytes and stromal cells. Recent studies have shown that BMSCs derived from mesoderm can also differentiate into neurons derived from ectoderm. On the basis of the successful selection of rat bone marrow mesenchymal stem cells by flow cytometry, a retinal / bone marrow mesenchymal stem cell co-culture system was used to simulate the microenvironment of retinal development in vivo. Bone marrow mesenchymal stem cells were induced to differentiate into retinal ganglion like cells. The differentiation level of the cells was identified by immunohistochemistry, neuronal specific antibody and flow cytometry, and the cDNA subtractive library was constructed by suppression subtractive hybridization before and after fine induction of bone marrow mesenchymal stem. To screen the differentially expressed genes during the differentiation of BMSCs into retinal ganglion like cells. The results showed that the differentially expressed genes GAP43 and RGS of bone marrow mesenchymal stem cells (BMSCs) were screened by SSH technique before and after induction. The results showed that BMSCs expressed some specific genes in retinal ganglion cells after induction. The results provide experimental basis for the study of the differentiation and function of BMSCs into retinal ganglion cells by transgenic method.
