【摘要】:目的:探討胃動素(Motilin,MTL)前體mRNA基因在甲狀腺組織中的表達及序列測定;觀察MTL活性肽在甲狀腺的定性表達和分布;檢測MTL及其mRNA在甲狀腺不同病變中的表達和臨床意義。 方法:采用RT-PCR、Southern Blot雜交和分子克隆等先進分子生物學技術(shù),克隆并測定人甲狀腺MTL前體mRNA基因序列;采用熒光免疫組織化學雙染技術(shù),觀察胃動素免疫活性物(MTL-IR)在甲狀腺組織中的分布和定性表達;采用實時熒光定量PCR(Real Time PCT,RT-PCR)和熒光免疫組化技術(shù),比較MTL前體mRNA和MTL活性肽在正常甲狀腺和甲狀腺良、惡性腫瘤組織中表達。 結(jié)果:(1)RT-PCR及Southern Blot雜交結(jié)果首次報道,人甲狀腺組織有MTL前體mRNA的表達。MTL mRNA分子序列測定結(jié)果顯示,人甲狀腺組織MTL mRNA基因序列與美國基因庫報道的人小腸MTL mRNA基因序列完全相同。(2)免疫熒光組化研究顯示,甲狀腺組織內(nèi)有大量MTL免疫陽性細胞的表達,該細胞主要分布在甲狀腺中央?yún)^(qū)域,散在于濾泡間或濾泡上皮之間,且MTL與降鈣素(Calcitonin,CT)共存,提示MTL表達于甲狀腺C細胞。(3)甲狀腺腫瘤組織內(nèi)有MTL免疫陽性物的表達,MTL免疫陽性細胞或彌散(髓樣癌)或集中(濾泡癌、乳頭癌、甲狀腺嗜酸性腺瘤)分布;甲狀腺髓樣癌和嗜酸性腺瘤的MTL表達明顯高于正常甲狀腺組織(P0.001);但與正常甲狀腺組織相比甲狀腺乳頭狀癌和濾泡癌MTL的表達明顯降低(P0.01);結(jié)節(jié)性甲狀腺腫與正常甲狀腺組織相比MTL的表達無顯著差異(P0.05)。(4)甲狀腺腫瘤組織內(nèi)有MTL前體mRNA的表達。甲狀腺髓樣癌和嗜酸性腺瘤的MTL前體mRNA的表達明顯高于正常甲狀腺組織(P0.01);但與正常甲狀腺組織相比甲狀腺乳頭狀癌和濾泡癌MTL前體mRNA的表達明顯降低(P0.05);結(jié)節(jié)性甲狀腺腫與正常甲狀腺組織相比MTLmRNA的表達無顯著差異(P0.05)。 結(jié)論:人甲狀腺組織有MTL前體mRNA的表達,且人甲狀腺組織MTL mRNA
[Abstract]:Objective: to investigate the expression and sequence determination of mRNA gene precursor of motilin motilinlus (MTL) in thyroid tissue, to observe the qualitative expression and distribution of MTL active peptide in thyroid gland, and to detect the expression and clinical significance of MTL and mRNA in different thyroid lesions. Methods: the precursor mRNA gene of human thyroid MTL was cloned and sequenced by RT-PCR Southern Blot hybridization and molecular cloning. To observe the distribution and qualitative expression of motilin immunoreactive substance (MTL-IRR) in thyroid tissue, to compare MTL precursor mRNA and MTL active peptide in normal thyroid and thyroid benign by real-time fluorescence quantitative PCR(Real Time PCT RT-PCR and fluorescence immunohistochemical technique. It is expressed in malignant tumor tissues. Results the results of RT-PCR and Southern Blot hybridization showed for the first time that MTL precursor mRNA was expressed in human thyroid tissue. The MTL mRNA gene sequence of human thyroid tissue was exactly the same as that of human small intestine MTL mRNA gene reported by American gene bank. The immunohistochemical study showed that a large number of MTL immunoreactive cells were expressed in thyroid tissue. The cells were mainly distributed in the central thyroid gland, scattered between follicles or between follicular epithelium, and MTL co-existed with calcitonin C +. It is suggested that the expression of MTL immunoreactive cells in thyroid neoplasms is expressed in thyroid C cells. 3) the distribution of MTL immunoreactive cells in thyroid neoplasms is diffuse (medullary carcinoma) or concentrated (follicular carcinoma, papillary carcinoma, thyroid eosinophilic adenoma). The expression of MTL in medullary thyroid carcinoma and eosinophilic adenoma was significantly higher than that in normal thyroid tissue (P 0.001), but the expression of MTL in thyroid papillary carcinoma and follicular carcinoma was significantly lower than that in normal thyroid tissue, and the expression of MTL in nodular goiter was significantly lower than that in normal thyroid tissue. There was no significant difference in the expression of MTL precursor mRNA in thyroid neoplasms compared with the expression of MTL. The expression of MTL precursor mRNA in medullary thyroid carcinoma and eosinophilic adenoma was significantly higher than that in normal thyroid tissue, but the expression of MTL precursor mRNA in thyroid papillary carcinoma and follicular carcinoma was significantly lower than that in normal thyroid tissue. There was no significant difference in the expression of MTLmRNA between adenoma and normal thyroid tissue. Conclusion: there is MTL precursor mRNA expression in human thyroid tissue and MTL mRNA in human thyroid tissue.
