抗原表位肽的快速篩選鑒定及其在免疫檢測和抗體制備中的應(yīng)用

【摘要】:抗原表位是抗原的核心部分,目前確定抗原表位的常用方法有:酶促或化學(xué)裂解片段分析法、蛋白印跡法、肽掃描、化學(xué)修飾或突變分析、表面呈現(xiàn)肽庫和隨機(jī)片段表達(dá)庫篩選,以及X射線衍射和核磁共振(NMR)分析等,這些方法大多操作繁瑣、難度較大,效率非常低。隨著計算機(jī)技術(shù)在生物技術(shù)領(lǐng)域的應(yīng)用和發(fā)展,研究發(fā)現(xiàn),抗原表位與蛋白質(zhì)的氨基酸序列的結(jié)構(gòu)特征存在特定的關(guān)系,從而建立了一系列基于這些結(jié)構(gòu)特征參數(shù)的抗原表位預(yù)測分析方案,預(yù)測抗原表位的正確率可以達(dá)到75%以上。在篩選鑒定抗原表位時,如果先通過抗原表位的預(yù)測使待選表位的范圍大大縮小,再用相應(yīng)的實驗來鑒定,往往可取得事半功倍的效果。
[Abstract]:Antigenic epitopes are the core of antigens. At present, the commonly used methods for the determination of antigen epitopes are enzymatic or chemical cleavage fragment analysis, Western blotting, peptide scanning, chemical modification or mutation analysis. Surface presentation peptide library and random fragment expression library screening, X-ray diffraction and nuclear magnetic resonance (NMR) analysis, most of these methods are cumbersome, difficult and inefficient. With the application and development of computer technology in the field of biotechnology, it has been found that there is a specific relationship between antigenic epitopes and the structural characteristics of amino acid sequences of proteins. A series of antigenic epitope prediction and analysis schemes based on these structural characteristic parameters are established, and the accuracy of antigen epitope prediction can reach more than 75%. In the screening and identification of antigen epitopes, if the range of epitopes to be selected is greatly reduced by the prediction of antigen epitopes, and then identified by corresponding experiments, the results can be doubled with half the effort.
