【摘要】:目的 隱孢子蟲(Cryptosporidium)是一種呈世界性分布的機會性致病原蟲,已被列為導致人體腹瀉的6種病原體之一。因其引起的水源性及食源性腹瀉的爆發(fā)性流行及可成為AIDS患者、免疫力低下兒童的致死性病因而倍受重視。國外已將隱孢子蟲作為AIDS患者必檢的項目之一。卵囊是隱孢子蟲主要的病原學診斷依據(jù)。不同動物源的隱孢子蟲卵囊在形態(tài)大小及致病性上有一定差異,因此隱孢子蟲卵囊的形態(tài)學研究對分類、致病性及流行病學研究具有一定的意義。有關隱孢子蟲卵囊的形態(tài)及大小已有一些描述,但至今尚未見采用數(shù)字圖像處理技術分析隱孢子蟲卵囊形態(tài)學參數(shù)的報道,也沒有見到用該技術對不同動物源隱孢子蟲卵囊進行對比分析的資料。與傳統(tǒng)的形態(tài)學研究方法相比,數(shù)字圖像處理技術可以更客觀、精確、靈活、大通量地獲取信息,且因具有快速、簡捷、易于操作等優(yōu)點而倍受青睞。近年來,該技術已被用于一些寄生蟲的形態(tài)學研究。本研究采用數(shù)字圖像分析技術建立并分析隱孢子蟲卵囊的形態(tài)學參數(shù),對比分析了沈陽地區(qū)小鼠源和長春地區(qū)牛源隱孢子蟲卵囊的形態(tài)大小,從數(shù)理分析角度確定了隱孢子蟲卵囊的形態(tài)學差異。本研究結(jié)果可為隱孢子蟲的病原學及流行病學研究提供更可靠的形態(tài)學資料。 材料與方法 1.隱孢子蟲卵囊的來源 1.1 沈陽地區(qū)KM小鼠源隱孢子蟲卵囊:中國醫(yī)科大學實驗動物部培育的昆明株(KM)小鼠,經(jīng)地塞米松免疫抑制處理,誘導小鼠隱孢子蟲感染,建立小鼠隱孢子蟲感染模型。取小鼠糞便經(jīng)涂片抗酸染色檢獲到隱孢
[Abstract]:Purpose Cryptosporidium (Cryptosporidium), a worldwide opportunistic protozoa, has been listed as one of the six pathogens leading to human diarrhea. Due to the outbreak of waterborne diarrhea and food-borne diarrhea, which can become the fatal cause of AIDS patients and children with low immunity, much attention has been paid to it. Cryptosporidium has been regarded as one of the necessary screening items for AIDS patients abroad. Oocysts are the main basis for etiological diagnosis of Cryptosporidium. The morphology and pathogenicity of Cryptosporidium oocysts from different animal sources are different, so the morphological study of Cryptosporidium oocysts has some significance for classification, pathogenicity and epidemiology. The morphology and size of Cryptosporidium oocysts have been described, but no digital image processing technique has been reported to analyze the morphological parameters of Cryptosporidium oocysts. There was no comparative analysis of Cryptosporidium oocysts from different animal sources by using this technique. Compared with the traditional morphological research methods, digital image processing technology can be more objective, accurate, flexible, large throughput of information acquisition, and has the advantages of fast, simple, easy to operate and so on. In recent years, this technique has been used to study the morphology of some parasites. In this study, the morphological parameters of Cryptosporidium oocysts were established and analyzed by digital image analysis, and the morphological size of Cryptosporidium oocysts in Shenyang and Changchun were compared. The morphological differences of Cryptosporidium oocysts were determined from the point of view of mathematical analysis. The results can provide more reliable morphological data for etiology and epidemiology of Cryptosporidium. Materials and methods 1. Source of Cryptosporidium oocysts 1.1 Cryptosporidium oocysts of km mice in Shenyang area: Kunming strain KM (Kunming strain KM) cultured in laboratory animal department of China Medical University was treated with dexamethasone immunosuppressive treatment to induce Cryptosporidium infection in mice and to establish the model of Cryptosporidium infection in mice.
