【摘要】:衰老是不可抗拒的自然規(guī)律,其發(fā)生發(fā)展是一個難解的謎團。近年來,我國逐步進入老齡化社會,有效地維護老年人的健康,防治衰老相關(guān)性疾病成為人們普遍關(guān)注的社會問題。因此,研究衰老發(fā)生機制及延緩衰老進程,也成為當今學術(shù)界倍受重視的領(lǐng)域。 關(guān)于衰老,人們從不同角度提出了多種學說,總的來說可以分為兩大類:一種認為衰老是人體固有的、隨時間退化的過程,稱為自然演進學說;另一種是認為衰老是各種內(nèi)外損傷積累的結(jié)果,稱為環(huán)境傷害學說。衰老的過程是內(nèi)外因素共同作用的結(jié)果,這兩類學說各自強調(diào)了其中一方面因素。近年來興起的衰老的線粒體-mtDNA學說認為,線粒體呼吸功能衰老和mtDNA結(jié)構(gòu)損傷之間的惡性循環(huán)是導致機體衰老的重要原因。此學說既肯定了遺傳物質(zhì)對衰老進程的重要作用,又沒有忽略環(huán)境損傷因素的影響,此學說一經(jīng)提出就得到了廣大學者的支持,成為當今衰老研究的一大熱點。 在衰老的研究中,合適的衰老動物模型對于深入研究衰老性疾病的發(fā)生機制、發(fā)展過程非常重要,也是抗衰老藥物的篩選、研制所必需的前提。近年來,中外學者通過給大鼠或小鼠注射大劑量D-半乳糖制作了亞急性衰老動物模型,并將此模型運用到衰老研究中,特別是抗衰老中藥的篩選研究。但是,學界對此動物模型的研究一直不夠深入,對其發(fā)生機理的闡述不夠明確。深入了解伴隨動物造模發(fā)生的一系列生化及分子生物學的改變,對于合理使用此種模型動物具有重大意義。衰老的線粒體-mtDNA學說的提出,為深入研究D-半乳糖致衰老動物模型提供了新的理論支持。 設(shè)計本課題,建立亞急性衰老動物模型。通過測定線粒體呼吸酶活性以觀察受試動物線粒體功能的衰老情況;通過測定MDA含量以及抗氧化酶
[Abstract]:Aging is an irresistible law of nature, its occurrence and development is a mystery. In recent years, China has gradually entered an aging society, effectively maintaining the health of the elderly and preventing and curing age-related diseases has become a social issue of universal concern. Therefore, the research on the mechanism of aging and the process of delaying aging has become an important field in academic circles. On aging, people put forward a variety of theories from different angles, in general can be divided into two categories: one is that aging is inherent in the human body, the process of degradation with time, called natural evolution theory; The other is that aging is the result of a variety of internal and external damage accumulation, called environmental injury theory. The process of aging is the result of internal and external factors, and the two theories emphasize one of them. In recent years, the mtDNA theory of aging suggests that the vicious circle between mitochondrial respiratory function senescence and mtDNA structural damage is an important cause of aging. This theory not only affirms the important role of genetic material in aging process, but also does not ignore the influence of environmental damage factors. As soon as this theory is put forward, it has been supported by many scholars and has become a hot spot in the field of aging research. In the research of aging, the suitable animal model of aging is very important for the further study of the mechanism of aging diseases and the development process. It is also the prerequisite for the screening of anti-aging drugs and the development of anti-aging drugs. In recent years, Chinese and foreign scholars have made subacute aging animal model by injecting large dose of D-galactose into rats or mice, and applied this model to the research of aging, especially the screening of anti-aging Chinese medicine. However, the research on the animal model has not been deep enough, and the mechanism of its occurrence is not clear enough. A deep understanding of a series of biochemical and molecular biological changes associated with animal modeling is of great significance for the rational use of this model animal. The mtDNA theory of aging provides a new theoretical support for the further study of aging animal model induced by D-galactose. Design this subject, establish the animal model of subacute senescence. The aging of mitochondria function was observed by measuring the activity of mitochondrial respiratory enzyme, and the content of MDA and antioxidant enzymes were measured.
