小鼠精原細(xì)胞分離培養(yǎng)及其影響因素的實(shí)驗(yàn)研究

【摘要】:據(jù)報(bào)道,近年來(lái)男性不育的發(fā)生率有所升高,無(wú)精、少精和弱精的患者有所增加,因而對(duì)精子發(fā)生機(jī)制及其影響因素的研究也就成了生殖醫(yī)學(xué)領(lǐng)域的一大研究熱點(diǎn)。精子發(fā)生從精原細(xì)胞開始,經(jīng)精母細(xì)胞和精子細(xì)胞,形成成熟的精子,其間經(jīng)歷多次有絲分裂和兩次減數(shù)分裂,受神經(jīng)、內(nèi)分泌、旁分泌及多種細(xì)胞因子的調(diào)節(jié),是一個(gè)極其復(fù)雜的生物學(xué)過程;對(duì)這一復(fù)雜過程及其影響因素的研究是預(yù)防和治療男性不育和研制男性節(jié)育方法的基礎(chǔ)性研究。目前這方面的研究多為在體研究,體外研究較少。在體研究結(jié)果顯示,導(dǎo)致男性精子數(shù)目及質(zhì)量下降的因素主要有遺傳缺陷、慢性疾患、過度勞累和精神負(fù)擔(dān)、環(huán)境污染、不良的飲食和生活習(xí)慣等。由于在體研究受神經(jīng)、體液等許多復(fù)雜因素的影響,因而在體外培養(yǎng)條件下研究生精過程及其影響因素便成了必然趨勢(shì)。 最原始的一種精原細(xì)胞即A型精原細(xì)胞是生精干細(xì)胞,通過非對(duì)稱性有絲分裂,這種細(xì)胞部分分化為B型精原細(xì)胞,另一部分則仍然保持為生精干細(xì)胞。在進(jìn)行體外研究生精過程的機(jī)制及其影響因素時(shí),最先遇到的一個(gè)問題就是如何提取和純化培養(yǎng)生精干細(xì)胞。本項(xiàng)研究以小鼠為實(shí)驗(yàn)動(dòng)物,首先研究了生精干細(xì)胞的提取、純化培養(yǎng)的最佳方法,然后檢測(cè)了溫度、EGF、重組卵泡刺激素、高氧等對(duì)生精干細(xì)胞存活、代謝、增殖、凋亡等的影響。 一.精原細(xì)胞的分離和純化培養(yǎng) 精原細(xì)胞中的A型精原細(xì)胞是一種生精干細(xì)胞,可通過非對(duì)稱性細(xì)胞分裂分化為B型精原細(xì)胞并進(jìn)行自我復(fù)制。目前,尚無(wú)法提取和純化這種精原干細(xì)胞,只能提取和純化培養(yǎng)含有生精干細(xì)胞的全部精原細(xì)胞。本實(shí)驗(yàn)篩選出了分離、培養(yǎng)和體外擴(kuò)增精原細(xì)胞的一種比較好的方法,為生精過程及其影響因素的體外研究提供了重要的技術(shù)支持。
[Abstract]:It is reported that the incidence of male infertility has increased in recent years, and the number of patients with azoospermia, oligozoospermia and asthenospermia has increased. Therefore, the research on the mechanism of spermatogenesis and its influencing factors has become a hot topic in the field of reproductive medicine. Spermatogenesis begins with spermatogonia and forms mature spermatozoa through spermatocytes and spermatocytes, which undergo multiple mitosis and two meiosis and are regulated by nerve, endocrine, paracrine and multiple cytokines. The study of this complex process and its influencing factors is the basic research on the prevention and treatment of male infertility and the development of male birth control methods. At present, most of the researches in this field are in vivo, but few in vitro. The results showed that the main factors leading to the decline of sperm number and quality were genetic defects, chronic diseases, overwork and mental burden, environmental pollution, poor diet and living habits, etc. Since in vivo research is influenced by many complicated factors such as nerve and body fluid, it is an inevitable trend for graduate students to study the process of spermatozoa and its influencing factors under the condition of culture in vitro. The most primitive spermatogonia, type A spermatogonial cells, are spermatogenic stem cells, which differentiate into B type spermatogonial cells by asymmetric mitosis, while the other part still maintain spermatogenic stem cells. In the process of in vitro spermatogenesis and its influencing factors, one of the first problems is how to extract and purify spermatogenic stem cells. In this study, mice were used as experimental animals. The best methods for extracting and purifying spermatogenic stem cells were studied, and then the survival, metabolism and proliferation of spermatogenic stem cells were detected, such as EGF, recombinant follicle stimulating hormone, hyperoxia, etc. The effect of apoptosis, etc. I. Isolation and purification of spermatogonia Type A spermatogonia in spermatogonia is a kind of spermatogenic stem cells, which can differentiate into B spermatogonia by asymmetric cell division and self-replicate. At present, it is not possible to extract and purify spermatogonial stem cells, but only to extract and purify all spermatogonia containing spermatogenic stem cells. A better method for isolation, culture and amplification of spermatogonia in vitro was screened out in this experiment. The in vitro study on the process of spermatogenesis and its influencing factors provided important technical support.
