β-欖香烯抑制膠質(zhì)瘤兔角膜移植模型血管生成的實(shí)驗(yàn)研究

【摘要】:目的復(fù)制兔角膜移植瘤血管生成模型,檢測(cè)β-欖香烯體內(nèi)抗血管生成效應(yīng)。方法裸鼠皮下種植人腦膠質(zhì)瘤細(xì)胞系SHG44細(xì)胞,形成實(shí)體膠質(zhì)瘤。將所得膠質(zhì)瘤實(shí)體小塊移植于兔角膜,復(fù)制角膜移植瘤血管生成模型。經(jīng)兔耳緣靜脈注射β-欖香烯,干預(yù)兩周后應(yīng)用Ⅷ因子相關(guān)抗原免疫組織化學(xué)染色及圖像分析技術(shù),檢測(cè)移植膠質(zhì)瘤組織內(nèi)微血管密度變化。結(jié)果兔角膜移植瘤血管生成模型成功復(fù)制;β-欖香烯處理組移植膠質(zhì)瘤微血管密度明顯低于對(duì)照組。結(jié)論兔角膜血管生成模型是定量研究腫瘤血管生成的良好工具;β-欖香烯具有體內(nèi)抗腫瘤血管生成作用。
[Abstract]:Objective to study the anti-angiogenesis effect of 尾-elemene in rabbit corneal transplantation tumor. Methods Human glioma cell line SHG44 was implanted subcutaneously in nude mice to form solid glioma. The grafts of glioma were transplanted into rabbit cornea and the angiogenesis model of corneal transplantation tumor was established. 尾 -elemene was injected intravenously through rabbit ear margin.
