巴馬長壽老人代謝相關(guān)基因GSTs、P450及抑癌基因P53的多態(tài)性研究

【摘要】: 目的:探討代謝相關(guān)基因GSTT1、GSTM1、CYP1A1及抑癌基因p53第4外顯子第72位密碼子(codon72)與人類長壽的關(guān)系。方法:應(yīng)用PCR-RFLP技術(shù)對(duì)巴馬156例(年齡90-105歲,平均93.2歲,長壽組)健康壯族長壽老人和142例健康壯族成年人(年齡20-72歲,平均34.8歲,對(duì)照組)的GSTT1、GSTM1、CYP1A1及p53基因進(jìn)行基因分型,了解這些基因的多態(tài)性,分析各種基因(型)頻率及多態(tài)性組合在長壽組和對(duì)照組之間的差異及其與長壽的關(guān)系。結(jié)果:長壽組的GSTT1基因、GSTM1基因的缺失(或攜帶)頻率、CYP1A1基因的野生型(T/T)、純合突變型(T/C)、雜合突變型(C/C)頻率與對(duì)照組差別均無顯著意義(p0.05);長壽組p53codon72片段的野生型(G/G)頻率明顯高于對(duì)照組(53%:29%,p0.05)。各種基因型的組合分析顯示,對(duì)照組同時(shí)合并GSTT1、GSTM1、CYP1A1及p53codon72基因缺失或者突變的幾率明顯比長壽組為高,尤其是長壽組男性和女性的GSTT1(+)-GSTM1(-)-CYP1A1 (T/C,C/C)- p53coden72(G/C,C/C)基因組合及長壽組女性GSTT1(-)-GSTM1(-)-CYP1A1 (T/C,C/C)-p53coden72 (G/C,C/C)基因組合明顯低于對(duì)照組( p0.05 );而長壽組男性GSTT1(-)-GSTM1(-)-CYP1A1 (T/C,C/C)-p53coden72 (G/G)基因組合高于對(duì)照組(p0.05),其它基因(型)組合在兩組中無差別。結(jié)論:GSTT1、GSTM1、CYP1A1及p53基因的多態(tài)性與人類長壽有著較為密切的聯(lián)系,其具體作用機(jī)制有待進(jìn)一步研究。
[Abstract]:Aim: to investigate the relationship between the metabolism-related gene GSTT1GSTM1CY CYP1A1 and codon 72of p53 exon 4. Methods: genotyping of CYP1A1 and p53 genes in 156 healthy adults (aged 20-72 years, mean 34.8 years, control group) from 156 healthy adults (90-105 years old, average 93.2 years old) and 142 healthy adults (20-72 years old, mean 34.8 years old, control group) was performed using PCR-RFLP technique. To understand the polymorphism of these genes, and to analyze the differences between longevity group and control group and their relationship with longevity. Results: there was no significant difference in the frequency of deletion (or carrier) of GSTT1 gene, wild type of CYP1A1 gene, homozygous mutant T / C, heterozygous mutation type C / C between the longevity group and the control group (p 0.05), while the wild type G / G) frequency of the p53codon72 fragment in the longevity group was not significantly different from that in the control group. The rate was significantly higher than that of the control group.
