大鼠燙傷后小腸粘膜T淋巴細(xì)胞的變化分析

【摘要】:目的了解TCRαβT 細(xì)胞和TCRγδT 細(xì)胞在正常大鼠小腸粘膜腸段間、上皮與固有層間的分布規(guī)律,探討它們?cè)跔C傷后的改變,以闡明燙傷后腸粘膜免疫功能的變化規(guī)律,為燙傷后腸源性感染的防治提供新的思路和形態(tài)學(xué)依據(jù)。 方法將40 只Wistar 雄性大鼠隨機(jī)分成4 組,即正常對(duì)照組、燙傷后1d 組、燙傷后3d 組和燙傷后7d 組,每組10 只。正常對(duì)照組,不給予蒸汽燙傷;另外三組用江西醫(yī)學(xué)院一附院燒傷實(shí)驗(yàn)中心研制的燙傷控制儀在背部脫毛區(qū)給予蒸汽燙傷,造成TBSA30%Ⅲo燙傷。經(jīng)抗休克處理后,分別于燙傷后1d(24h)、3d(72h)和7d 剖腹取十二指腸、空腸和回腸。恒冷切片機(jī)切片,對(duì)相鄰切片用小鼠抗大鼠CD_3單克隆抗體和小鼠抗大鼠TCRγδ單克隆抗體ABC免疫組織化學(xué)方法顯示CD_3~+T 細(xì)胞和TCRγδT 細(xì)胞,用方網(wǎng)測(cè)試系統(tǒng)測(cè)量和計(jì)數(shù),根據(jù)測(cè)量差值求出TCRαβT 細(xì)胞的數(shù)據(jù),分別求出其體密度、數(shù)密度、表面積密度、平均體積和表面積體積比等體視學(xué)參數(shù)。用統(tǒng)計(jì)軟件SPSS 10.0 for window 比較它們?cè)谀c段間、上皮與固有層間的差異。 結(jié)果(1)正常(未燙傷)大鼠十二指腸和空腸上皮內(nèi)TCRγδT 細(xì)胞數(shù)密度比TCRαβT 細(xì)胞大,而回腸上皮內(nèi)TCRγδT 細(xì)胞數(shù)密度比TCRαβT 細(xì)胞小。從十二指腸到回腸,上皮內(nèi)CD_3~+T 細(xì)胞和TCRγδT 細(xì)胞數(shù)密度依次遞減,而TCRαβT 細(xì)胞數(shù)密度遞增。(2)各腸段固有層TCRαβT 細(xì)胞數(shù)密度比TCRγδT 細(xì)胞大。從十二指腸到回腸,固有層中CD_3~+T 細(xì)胞和TCRαβT 細(xì)胞的數(shù)密度依次遞增,而TCRγδT 細(xì)胞的數(shù)密度變化不大。(3)十二指腸和空腸上皮內(nèi)CD3+T 細(xì)胞和TCRγδT 細(xì)胞數(shù)密度比固有層中的大;而回腸上皮內(nèi)CD3+T 細(xì)胞和TCRγδT 細(xì)胞數(shù)密度比固有層中的小。各腸段上皮內(nèi)TCRαβT 細(xì)胞數(shù)密度比固有層中的小。(4)各腸段粘膜中TCRγδT 細(xì)胞平均體積比TCRαβT 細(xì)胞的大。上皮內(nèi)T 細(xì)胞的平均體積比固有層T 細(xì)胞的大。各腸段間無(wú)差異。(5)上皮內(nèi)T 細(xì)胞表面積體積比值比固有層T 細(xì)胞的大,各腸段間無(wú)差異。TCRαβT 細(xì)胞與TCRγδT 細(xì)胞間也無(wú)差異。(6)燙傷后各腸段上皮和固有層CD3+T
