耐特定抗菌藥雙歧桿菌的鑒定與特性研究

【摘要】: 本項(xiàng)研究以源自內(nèi)蒙古呼和浩特嬰兒及牛犢的10株雙歧桿菌為試材,用傳統(tǒng)鑒定法(氣相色譜法和糖發(fā)酵法),進(jìn)行雙歧桿菌的屬、種鑒定,同時(shí)用引物(Eu-01、Eu-02,Bif-03、Bif-04)對(duì)受試菌株DNA進(jìn)行擴(kuò)增,在電泳圖上,各菌株均擴(kuò)增出了預(yù)期大小的多重PCR產(chǎn)物,說(shuō)明該P(yáng)CR方法可將雙歧桿菌鑒定到屬水平,且結(jié)果穩(wěn)定,特異性好。 對(duì)雙歧桿菌馴化后的耐氧菌株、實(shí)驗(yàn)的原始菌株以及傳代30-40次以后的厭氧菌株的生長(zhǎng)特性及其對(duì)頭孢氨芐(cefalexin)、阿米卡星(Amikacin)、甲硝唑(Metronidazole)三種抗菌藥的耐藥性等特性進(jìn)行了研究比較;同時(shí)研究了雙歧桿菌的質(zhì)粒與耐藥性的關(guān)系。結(jié)果表明: 隨著傳代次數(shù)的增加,2株豚雙歧桿菌由原來(lái)對(duì)頭孢氨芐的耐藥變?yōu)橹卸让舾?對(duì)其它兩種抗生素仍然為耐藥;雖然其它菌株的耐藥性未發(fā)生改變,但耐藥強(qiáng)度有所減弱,即對(duì)同種抗菌藥同一濃度的抑菌圈直徑增大,且差異顯著(p0.05)。 需氧馴化后雙歧桿菌的耐藥性變化不大,只是個(gè)體形態(tài)均變?yōu)槎虠U狀或球狀,生長(zhǎng)速度減緩。 采用堿裂解法提取并檢測(cè)了雙歧桿菌的耐藥性質(zhì)粒,僅在一株標(biāo)準(zhǔn)雙歧桿菌B2778中發(fā)現(xiàn)有1條質(zhì)粒帶,其對(duì)應(yīng)的核酸分子量大約是23kbps,其他受試菌株中未發(fā)現(xiàn)有質(zhì)粒存在,從而表明這些菌株對(duì)多種抗菌藥的耐藥性并非由質(zhì)粒介導(dǎo),這些菌株不存在耐藥因子傳遞的危險(xiǎn)。
[Abstract]:In this study, 10 strains of Bifidobacterium from infants and calves in Hohhot, Inner Mongolia, were used to identify the genus and species of Bifidobacterium by traditional identification methods (gas chromatography and sugar fermentation). At the same time, the DNA of the tested strain was amplified by primer Eu-01and Eu-02Bif-03BIF-04. On the electrophoretic map, the expected size of multiple PCR products were amplified from each strain, indicating that the PCR method could identify Bifidobacterium to the generic level, and the results were stable and specific. After acclimation of Bifidobacterium, the growth characteristics of the original strain and the anaerobic strain after 30 to 40 times of passage were studied and compared with those of the three antimicrobial agents, such as cefalexin, Amikacinine, Metronidazole. etc. The relationship between plasmid and drug resistance of Bifidobacterium was also studied. The results show that: With the increase of the number of passages, two strains of bifidobacterium changed from resistance to cefalexin to moderate sensitivity, and remained resistant to the other two antibiotics. Although the resistance of other strains remained unchanged, the drug resistance of the other strains weakened. The diameter of bacteriostasis circle of the same concentration of the same antimicrobial drug increased, and the difference was significant (p 0.05). After aerobic acclimation, the drug resistance of Bifidobacterium changed little, but the individual morphology became short rod or globular, and the growth rate slowed down. The resistant plasmids of Bifidobacterium were extracted and detected by alkaline lysis method. Only one plasmid band was found in one standard strain B2778. The molecular weight of nucleic acid was about 23kbps. no plasmids were found in other strains. The results showed that the resistance of these strains to various antimicrobial agents was not mediated by plasmids, and there was no risk of drug resistance factor transmission in these strains.
