【摘要】:睪丸的精子發(fā)生是一個(gè)獨(dú)特復(fù)雜的細(xì)胞分裂分化過程。睪丸生精小管中的生精細(xì)胞先后經(jīng)過三個(gè)階段的變化:精原細(xì)胞的增殖,精母細(xì)胞的減數(shù)分裂和精子細(xì)胞的變態(tài)過程,最終產(chǎn)生成熟的精子。許多在睪丸中特異或高度表達(dá)的基因參與這一過程?寺〔G丸精子發(fā)生相關(guān)基因并研究其功能,對(duì)于揭示精子發(fā)生過程中減數(shù)分裂和變態(tài)的分子機(jī)制,對(duì)于臨床男性不育的診斷和治療以及男性分子節(jié)育等均具有重要的意義。 本實(shí)驗(yàn)室先期利用人睪丸cDNA microarray雜交技術(shù)比較了人胚胎期睪丸和成年期睪丸的基因表達(dá)差異,篩選出386個(gè)差異表達(dá)基因,ALF是其中的一條睪丸特異表達(dá)的基因,它是通用轉(zhuǎn)錄因子TFⅡA大亞單位(TFⅡAα/β)的類似物,,是生殖細(xì)胞特異形式的轉(zhuǎn)錄因子。本研究著重于ALF在人精子發(fā)生中的作用,以及其表達(dá)水平的改變與男性生育之間的關(guān)系。cDNA芯片雜交結(jié)果顯示ALF高表達(dá)于成人睪丸組織,在胚胎睪丸中表達(dá)量較低;16例生精障礙患者睪丸的RT-PCR結(jié)果顯示:ALF在2例唯支持細(xì)胞綜合癥病人睪丸中表達(dá)缺失;在7例生精阻滯患者睪丸中,有一例沒有表達(dá),另外6例有強(qiáng)弱不等的表達(dá);在7例生精紊亂患者睪丸中,1例表達(dá)缺失。原核表達(dá)ALF重組蛋白,純化并免疫小鼠制備多克隆抗體,經(jīng)Western Blot分析顯示ALF抗體在人睪丸和精子中均顯示有特異條帶,其分
[Abstract]:Testicular spermatogenesis is a unique and complex process of cell division and differentiation. Spermatogenic cells in testicular seminiferous tubules undergo three stages: spermatogonia proliferation, spermatocyte meiosis and sperm cell metamorphosis, resulting in mature spermatozoa. Many genes that are specific or highly expressed in the testis are involved in this process. Cloning testicular spermatogenesis related genes and studying their functions are of great significance in revealing the molecular mechanism of meiosis and metamorphosis during spermatogenesis, in the diagnosis and treatment of clinical male infertility and in male molecular sterility. The difference of gene expression between human embryonic testis and adult testis was compared by using human testis cDNA microarray hybridization technique, and 386 differentially expressed genes were screened out.
