【摘要】： 目的建立人食管癌免疫SCID小鼠異位移植模型并對其轉移性淋巴來源細胞特性進行初級研究。方法將食管癌細胞系Eca-109細胞懸液注射至SCID小鼠胃壁。三個月后或動物頻死時處死解剖,觀察腫瘤在小鼠體內的生長轉移情況,將肉眼可見的縱膈淋巴結轉移瘤塊接種于SCID鼠皮下擴增,然后取小鼠皮下瘤組織進行細胞培養(yǎng),得到穩(wěn)定的細胞株后用MTT法分析細胞的生長曲線,Western bloting法檢測拓撲異構酶的表達,酶譜法檢測MMP-2,MMP-9的活性,劃痕實驗和Transwell體外移動實驗檢測細胞的移動能力。結果成功建立裸鼠胃壁篩選第一代高轉移潛力細胞,并命名為NMC109,對其生物學特征研究發(fā)現其細胞增殖能力,移動能力和侵襲潛力都發(fā)生明顯增強。結論:腫瘤胃壁移植是建立食管癌免疫缺陷動物自發(fā)性移植模型和篩選轉移性細胞亞系的可行方法。
[Abstract]:Objective to establish the heterotopic transplantation model of human esophageal carcinoma immunized SCID mice and to study the characteristics of metastatic lymphoid cells.Methods the suspension of esophageal carcinoma cell line Eca-109 was injected into the gastric wall of SCID mice.After three months or when the animal died frequently, the tumor was killed and the tumor growth and metastasis were observed in mice. The mediastinal lymph node metastases were inoculated into SCID mice subcutaneously, and then the subcutaneous tumor tissues were taken from the mice for cell culture.After the stable cell line was obtained, the expression of topoisomerase was detected by Western bloting, the activity of MMP-2 and MMP-9 was detected by MTT assay, and the mobility of cells was detected by scratch test and Transwell in vitro mobility assay.Results the first generation of high metastatic potential cells were successfully established in nude mice gastric wall and named NMC109. The biological characteristics of NMC109 showed that the cell proliferation, mobility and invasion potential of NMC109 cells were significantly enhanced.Conclusion: tumor gastric wall transplantation is a feasible method for the establishment of spontaneous transplantation model and screening of metastatic cell sublines in immunodeficient esophageal carcinoma.
