【摘要】：精子發(fā)生是一個(gè)連續(xù)的細(xì)胞分裂分化過(guò)程，包括精原細(xì)胞分裂增殖、精母細(xì)胞減數(shù)分裂和精子形成三個(gè)主要階段。精子發(fā)生過(guò)程涉及染色體數(shù)目的減半、遺傳重組、染色質(zhì)重塑及濃縮、精子頂體和尾部的形成等復(fù)雜的變化過(guò)程，所有這些高度有序的過(guò)程均需要相關(guān)蛋白質(zhì)進(jìn)行精確地調(diào)節(jié)。分離和鑒定與精子發(fā)生相關(guān)的蛋白質(zhì)并探討其相關(guān)功能對(duì)于揭示男性生殖的分子調(diào)控機(jī)制及臨床男性不育癥的診治具有重要的理論和實(shí)踐意義。 為了研究小鼠睪丸局部短暫加熱導(dǎo)致小鼠生精障礙的分子機(jī)制，我們運(yùn)用蛋白質(zhì)組學(xué)研究常用的技術(shù)手段—雙向凝膠電泳(two-dimensional electrophoresis，2-DE)和質(zhì)譜(mass spectrometry，MS)分離和鑒定小鼠睪丸加熱后發(fā)生改變的蛋白質(zhì)。首先我們創(chuàng)建了含有小鼠睪丸雙向凝膠電泳參考膠的符合聯(lián)盟標(biāo)準(zhǔn)的數(shù)據(jù)庫(kù)，在參考膠上有504個(gè)通過(guò)肽質(zhì)量指紋譜鑒定的蛋白點(diǎn)，點(diǎn)擊膠上的蛋白點(diǎn)可鏈接到對(duì)應(yīng)的蛋白質(zhì)條目，同時(shí)還可得到蛋白質(zhì)的鑒定信息，并可鏈接到SWISS-PROT或NCBI蛋白質(zhì)網(wǎng)絡(luò)數(shù)據(jù)庫(kù)，另外，數(shù)據(jù)庫(kù)支持復(fù)雜的查詢功能，該公共數(shù)據(jù)庫(kù)為研究正常小鼠睪丸功能和小鼠睪丸病理提供了有價(jià)值的資源；其次，我們采用類似的方法分析了小鼠睪丸局部
[Abstract]:Spermatogenesis is a continuous process of cell division and differentiation, which consists of three main stages: spermatogonia division and proliferation, spermatocyte meiosis and spermatogenesis.Spermatogenesis involves complex processes such as halving the number of chromosomes, genetic recombination, chromatin remodeling and condensation, and the formation of acrosome and caudal sperm. All these highly ordered processes need to be precisely regulated by related proteins.The isolation and identification of proteins associated with spermatogenesis and their related functions are of great theoretical and practical significance in revealing the molecular regulatory mechanisms of male reproduction and the diagnosis and treatment of clinical male infertility.In order to study the molecular mechanism of mouse spermatogenic disorder caused by local transient heating of testis in mice,Two dimensional electrophoresis2-DEE and MS were used to isolate and identify the proteins changed after testicular heating in mice by means of proteomics, two dimensional electrophoretic electrophoresis (2-DE) and mass mass spectroscopy (MSM).First of all, we created a database containing mouse testis two-dimensional gel electrophoresis reference gel that meets the alliance standard. There are 504 protein spots identified by peptide mass fingerprinting on the reference gel.Clicking on the protein dot on the glue links to the corresponding protein entry, and you can also get the protein identification information and link to the SWISS-PROT or NCBI protein network database. In addition, the database supports complex query functions.The common database provides valuable resources for the study of testicular function and testicular pathology in normal mice. Secondly, we used a similar method to analyze the local testis of mice.
