【摘要】:睫狀神經(jīng)營養(yǎng)因子(CNTF)是分子量為22-26KD的酸性蛋白質(zhì)。因為最初是從雞的睫狀體中提取出來,并可維持雞副交感神經(jīng)節(jié)的存活而得名。它與NTF(neurotrophic factor)家族明顯不同,,沒有同源性,是非靶源性的營養(yǎng)因子,與白血病抑制因子(LIF)、白細胞介素6(IL-6)等具有相似的螺旋框架結(jié)構(gòu),因而被認為屬于IL-6家族。CNTF具有多種功能,能促使多種神經(jīng)細胞的存活,是第一個被發(fā)現(xiàn)的能維持在體和離體脊髓運動神經(jīng)元的存活及突起生長的神經(jīng)營養(yǎng)因子,CNTF還具有肌肉營養(yǎng)作用,在神經(jīng)系統(tǒng)發(fā)育、分化和神經(jīng)損傷修復中具有非常重要的作用。最近研究表明,睫狀神經(jīng)營養(yǎng)因子與位于丘腦部位的受體結(jié)合,能使缺乏leptin的肥胖小鼠(ob/ob mice)脂肪減少,體重降低;對于更能代表人類肥胖癥實際情況的食物誘導肥胖小鼠(diet-induced obesity,DIO)模型,CNTF也能使之體重下降。與通過飲食減肥及其他的治療方法相比,CNTF不會發(fā)生停藥后過量攝入食物而引起的體重反彈。因此,CNTF具有廣闊的臨床開發(fā)前景,有望開發(fā)成為一種新型、高效的減肥產(chǎn)品。 CNTF受體在神經(jīng)系統(tǒng)和骨骼肌中廣泛存在,在運動神經(jīng)系統(tǒng)以及與運動系統(tǒng)有關(guān)的部位大量表達。CNTF受體由CNTFRα、gp130、LIFRβ三部份組成,其中CNTFRα是CNTF特異性結(jié)合部位,決定哪些細胞對CNTF有反應,而gp130、LIFRβ與信號傳導有關(guān)。CNTF的諸多生物學功能主要是通過激活CNTFRα,然后介導信號轉(zhuǎn)導完成的。因此,通過對CNTF與CNTFRα相互作用的分子機制的研究,確定CNTF的關(guān)鍵
[Abstract]:Ciliary neurotrophic factor (CNTF) is an acidic protein with molecular weight of 22-26KD.It was originally extracted from the ciliary body of the chicken and can maintain the survival of the parasympathetic ganglion.It is different from NTF(neurotrophic family and has no homology. It is a non-target source nutrition factor, and has similar helical frame structure with leukemia suppressor, interleukin-6 (IL-6) and so on. Therefore, it is considered to belong to the IL-6 family. CNTF has many functions.CNTF, the first neurotrophic factor found to maintain the survival and growth of motor neurons in vivo and in vitro, also has muscular nutrition and develops in the nervous system.Differentiation and repair of nerve injury play a very important role.Recent studies have shown that ciliary neurotrophic factors combined with receptors located in the thalamus can reduce fat and weight in obese mice lacking leptin.The CNTF can also make the obese mice lose weight, which is more representative of the actual situation of human obesity.Compared with diet and other treatments, CNTF does not cause weight rebound due to excessive intake of food after withdrawal.Therefore, CNTF has a broad prospect of clinical development and is expected to become a new and efficient weight loss product.CNTF receptors are widely expressed in the nervous system and skeletal muscle. CNTF-receptors are highly expressed in the motor nervous system and motor system-related sites. CNTF-receptors are composed of three parts of CNTFR 偽 gp130 LIFR 尾, in which CNTFR 偽 is a specific binding site for CNTF, which determines which cells respond to CNTF.However, many biological functions of gp130 LIFR 尾 related to signal transduction are mainly accomplished by activating CNTFR 偽 and mediating signal transduction.Therefore, by studying the molecular mechanism of the interaction between CNTF and CNTFR 偽, the key of CNTF is determined.
