染氟成纖維細(xì)胞和成骨細(xì)胞IGF-1及bFGF的表達(dá)及其意義

【摘要】: 成纖維細(xì)胞(fibroblast,FB)屬于誘導(dǎo)性骨祖細(xì)胞范疇,在一定誘導(dǎo)因素的作用下具有向成骨細(xì)胞(osteoblast,OB)轉(zhuǎn)化并發(fā)揮成骨功能的特點(diǎn)。IGF-1、bFGF與骨生長代謝密切相關(guān),是骨代謝調(diào)控網(wǎng)絡(luò)中的重要細(xì)胞因子,二者在促進(jìn)FB和OB分化和增殖等方面有著重要的作用。本文將通過觀察氟化物對FB和OB中IGF-1、bFGF表達(dá)水平的影響,探討氟化物對OB成骨功能的促進(jìn)作用和對FB成骨功能的誘導(dǎo)增強(qiáng)效應(yīng),并討論這些作用在慢性氟中毒所致骨周軟組織骨化和鈣化的發(fā)生機(jī)制中的重要意義。 以小鼠成纖維細(xì)胞株和小鼠顱骨成骨樣細(xì)胞為實(shí)驗(yàn)對象,應(yīng)用ELISA、免疫組化等方法,檢測染氟FB和OB中IGF-1、bFGF蛋白的表達(dá)。結(jié)果表明,氟化物對FB和OB中IGF-1、bFGF蛋白的表達(dá)增強(qiáng)作用,在時間段和所需氟劑量上存在差異,但是具有相同的變化趨勢。RT-PCR檢查結(jié)果表明染氟FB中的IGF-1和bFGF mRNA表達(dá)增強(qiáng)的趨勢明顯。而且,IGF-1和bFGF對染氟后表達(dá)改變的時間段上存在一定的差異,提示二者在促進(jìn)FB成骨功能增強(qiáng)方面可能具有互補(bǔ)作用。 在單層細(xì)胞培養(yǎng)的基礎(chǔ)上,建立三維培養(yǎng)體系,研究在更接近體內(nèi)環(huán)境條件下,染氟FB中IGF-1蛋白和mRNA的表達(dá)及bFGF mRNA的表達(dá)。結(jié)果表明在兩種培養(yǎng)體系中,染氟FB在IGF-1和bFGF表達(dá)增強(qiáng)的程度上存在一定的差異,但變化趨勢相似,為證明氟化物對FB成骨功能的促進(jìn)增強(qiáng)效應(yīng)提供了進(jìn)一步的佐證。
[Abstract]:Fibroblast fibroblast (FB) belongs to the category of induced osteoblast progenitor cells. It has the characteristics of transforming to osteoblastus OBB and exerting osteogenic function. IGF-1bFGF is closely related to bone growth and metabolism.They play an important role in promoting the differentiation and proliferation of FB and OB.By observing the effect of fluoride on the expression of IGF-1bFGF in FB and OB, the effects of fluoride on the osteogenic function of OB and the induction and enhancement of osteogenic function of FB were studied.The significance of these roles in the pathogenesis of ossification and calcification of periosseous soft tissue caused by chronic fluorosis was discussed.The expression of IGF-1bFGF in fluorinated FB (FB) and OB (OB) was detected by ELISA-immunocytochemistry in mouse fibroblasts and osteoblast-like cells.The results showed that fluoride could enhance the expression of IGF-1bFGF protein in FB and OB, but had the same change trend. RT-PCR results showed that the expression of IGF-1 and bFGF mRNA in FB exposed to fluoride was significantly increased.Moreover, there were some differences between IGF-1 and bFGF on the expression of FB after fluoride exposure, suggesting that they might play a complementary role in enhancing the osteogenic function of FB.On the basis of monolayer cell culture, a three-dimensional culture system was established to study the expression of IGF-1 protein and mRNA and the expression of bFGF mRNA in fluorinated FB.The results showed that in the two culture systems, there were some differences in the enhancement of FB expression in IGF-1 and bFGF, but the change trend was similar, which provided further evidence for the enhancement effect of fluoride on the osteogenic function of FB.
