【摘要】:人類疾病往往是環(huán)境因素與遺傳因素共同作用的結(jié)果,探討基因與環(huán)境的相互作用機(jī)制與規(guī)律,對(duì)于防治環(huán)境因素造成的機(jī)體危害具有重要意義。 1998年,周建偉等利用差異顯示高通量篩選技術(shù)從培養(yǎng)的原代人的氣管和支氣管上皮細(xì)胞中分離克隆了一個(gè)在生物進(jìn)化上高度保守的環(huán)境應(yīng)答基因-JWA,其編碼蛋白是一種新的微管相關(guān)蛋白,并受多種分化誘導(dǎo)劑如氯化高鐵血紅素(hemin)等調(diào)控。該基因可能參與多種白血病細(xì)胞的定向分化和凋亡,并與細(xì)胞應(yīng)答環(huán)境中的熱應(yīng)激的信號(hào)轉(zhuǎn)導(dǎo)通路有關(guān)。 為了進(jìn)一步深入了解JWA基因與紅白血病的關(guān)系,本課題用hemin、熱應(yīng)激及hemin與熱應(yīng)激聯(lián)合處理K562細(xì)胞,觀察JWA基因在這些細(xì)胞模型中表達(dá)的特點(diǎn)和規(guī)律,探討JWA在細(xì)胞應(yīng)答hemin和應(yīng)激信號(hào)通路中的作用和分子機(jī)制,以期為發(fā)展新的物理療法與化療結(jié)合治療紅白血病提供依據(jù)。 一、JWA對(duì)hemin處理K562細(xì)胞的應(yīng)答:
[Abstract]:Human disease is often the result of the interaction of environmental factors and genetic factors. It is of great significance to explore the interaction mechanism and law of genes and environment for the prevention and treatment of environmental hazards.In 1998,Zhou Jianwei et al used differential display high-throughput screening technique to isolate and clone a highly conserved environmental response gene, -JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA-JWA, a highly conserved environmental response gene, was isolated from cultured primary human tracheobronchialMicrotubule-associated proteins,It is regulated by many differentiation inducers such as hemin.This gene may be involved in the directional differentiation and apoptosis of various leukemic cells and may be related to the signal transduction pathway of heat stress in the cellular response environment.In order to further understand the relationship between JWA gene and erythroleukemia, K562 cells were treated with hemin, heat stress, hemin and heat stress, and the characteristics and regularity of JWA gene expression in these cell models were observed.To explore the role and molecular mechanism of JWA in cellular response to hemin and stress signaling pathway in order to provide evidence for the development of new physiotherapy combined with chemotherapy in the treatment of erythroleukemia.The response of JWA to K562 cells treated with hemin:
