【摘要】：FCR1基因是白念珠菌鋅簇轉(zhuǎn)錄因子,它能夠負(fù)向調(diào)控白念珠菌的多藥耐藥現(xiàn)象,但其調(diào)控的分子機(jī)制目前尚不清楚。本課題利用藥物敏感性實(shí)驗(yàn)、羅丹明6G外排實(shí)驗(yàn)、薄層色譜分析、RT-PCR、Realtime-PCR等方法研究白念珠菌鋅簇轉(zhuǎn)錄因子FCR1參與多藥耐藥的分子機(jī)制,證明FCR1基因在氟康唑誘導(dǎo)的條件下能夠負(fù)向調(diào)控多藥耐藥基因CDR1、CDR2和MDR1的表達(dá),增強(qiáng)白念珠菌的藥物外排功能,從而使白念珠菌產(chǎn)生抗藥性。另外,基因芯片雜交技術(shù)發(fā)現(xiàn)FCR1缺失以后不同功能分類的基因表達(dá)差異,說(shuō)明FCR1基因可能參與白念珠菌多種生命活動(dòng)。其中菌絲和毒力相關(guān)基因NAG2和CTA1表達(dá)上調(diào),RPL6基因表達(dá)下調(diào),通過(guò)菌絲誘導(dǎo)和毒力實(shí)驗(yàn)證明FCR1缺失能夠負(fù)向調(diào)控白念珠菌菌絲的生長(zhǎng)和毒力。
[Abstract]:FCR1 gene is a zinc cluster transcription factor of Candida albicans, which can negatively regulate the multidrug resistance of Candida albicans, but its molecular mechanism is still unclear.In this study, drug sensitivity test, Rhodamine 6G efflux test and TLC analysis were used to study the molecular mechanism of zinc cluster transcription factor (FCR1) in Candida albicans involved in multidrug resistance.It is demonstrated that FCR1 gene can negatively regulate the expression of CDR1, CDR2 and MDR1 under fluconazole-induced condition, and enhance the drug efflux function of Candida albicans, thus causing Candida albicans to develop drug resistance.In addition, the gene expression of different functional classifications after FCR1 deletion was found by gene chip hybridization, indicating that FCR1 gene may be involved in many kinds of life activities of Candida albicans.Among them, hyphal and virulence related genes NAG2 and CTA1 up-regulated the expression of RPL6 gene. It was proved by hyphal induction and virulence experiment that FCR1 deficiency could negatively regulate the growth and virulence of Candida albicans.
