惡性瘧原蟲抗原對(duì)健康人外周血T細(xì)胞免疫功能的影響

【摘要】:目的探討惡性瘧原蟲抗原對(duì)健康人外周血T淋巴細(xì)胞免疫功能的影響。方法健康成人外周血單個(gè)核細(xì)胞(PBMC)體外分別用惡性瘧原蟲(P.f)抗原(5μg/ml)和正常紅細(xì)胞(nRBC)抗原(5μg/ml)進(jìn)行刺激,同時(shí)給予IL-2維持細(xì)胞增殖,另外設(shè)立只加IL-2的陰性對(duì)照組進(jìn)行培養(yǎng)。培養(yǎng)12 d時(shí),刺激組細(xì)胞再用對(duì)應(yīng)的抗原刺激20 h,用流式細(xì)胞儀檢測(cè)T淋巴細(xì)胞亞群分泌IL-4和IFN-γ的情況,并用羧基熒光素乙酰乙酸琥珀酰亞胺酯(Carboxyfluorescein diacetate,succinimidyl ester,CFSE)標(biāo)記法檢測(cè)T細(xì)胞增殖反應(yīng)。結(jié)果健康人PBMC經(jīng)P.f抗原刺激擴(kuò)增后CD4+T細(xì)胞的增殖指數(shù)(PI)明顯高于nRBC抗原刺激組和陰性對(duì)照組(P均0.05),但3組γδT的PI差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。P.f抗原組分泌IL-4的CD4+T細(xì)胞百分率明顯高于nRBC抗原組和陰性對(duì)照組(P均0.05),但3組分泌IFN-γ的CD4+T細(xì)胞百分率差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 P.f抗原在體外可刺激健康人外周血CD4+T細(xì)胞增殖活化,后者通過(guò)優(yōu)先分泌IL-4而發(fā)揮免疫調(diào)節(jié)作用。
[Abstract]:Objective to investigate the effect of Plasmodium falciparum antigen on the immune function of peripheral blood T lymphocytes in healthy adults. 5 渭 g / ml) and 5 渭 g / ml of normal RBC antigen were stimulated. At the same time, IL-2 was given to maintain cell proliferation, and a negative control group with only IL-2 was set up for culture. After 12 days of culture, the cells in the stimulation group were stimulated with the corresponding antigen for 20 h. It was significantly higher than that of nRBC antigen stimulation group and negative control group (P 0.05). However, the Pi difference of 緯 未 T in the three groups was not statistically significant. The percentage of CD4 T cells secreting IL-4 in the P0.05 + P.F antigen group was significantly higher than that in the nRBC antigen group and the negative control group (P < 0.05). P = 0.05). However, there was no significant difference in the percentage of CD4 T cells secreting IFN- 緯 among the three groups. Conclusion P.F antigen can stimulate the proliferation and activation of CD4 T cells in healthy human peripheral blood in vitro. The latter plays an immunomodulatory role by preferentially secreting IL-4.
