【摘要】:骨形成蛋白(Bone Morphogenetic Proteins,BMPs)是一種分泌型糖蛋白,具有多種生物學(xué)功能。BMP2和BMP4是骨形成蛋白家族中的兩個重要成員,在結(jié)構(gòu)和功能上這兩種蛋白都非常相似,兩者都能夠誘導(dǎo)骨組織的生成和造血組織的分化等,但BMP2誘導(dǎo)骨組織形成的活性更強,BMP4則對造血組織的作用更突出。在成體,BMP2誘導(dǎo)異位骨形成,成為用于骨組織工程的重要誘導(dǎo)因子:BMP4則可以促進造血干細胞的增殖,并誘導(dǎo)造血干細胞向紅系、粒系和巨噬細胞系三系細胞分化,從而能夠促進受損傷的造血系統(tǒng)的全面再生與恢復(fù);這些特性使得BMP2和BMP4都具有很好的臨床應(yīng)用前景。 在BMP的制備過程中,真核表達系統(tǒng)表達量低,表達產(chǎn)物回收、純化困難、生產(chǎn)成本很高;原核表達系統(tǒng),特別是大腸桿菌系統(tǒng),具有表達效率高,容易操作,容易監(jiān)控,產(chǎn)物穩(wěn)定,生產(chǎn)成本低等優(yōu)點,故而我們在進行BMP的大規(guī)模制備過程中選擇了大腸桿菌表達系統(tǒng)。目前,重組人骨形成蛋白2的大規(guī)模制備已有較多報道,,本研究的目的,就是應(yīng)用我們目前已經(jīng)比較成熟的制備BMP2的工藝,來制備BMP4,來驗證大規(guī)模制備這兩種蛋白時,能否用相同的發(fā)酵程序及相近的純化方法。 兩種工程菌株(DH5α/pDH2-BMP-4m及DH5α/pDH2-BMP-2m)分別
[Abstract]:Bone morphogenetic protein (BMP) is a secretory glycoprotein. BMP2 and BMP4 are two important members of the bone morphogenetic protein family. They are very similar in structure and function. Both of them could induce bone tissue formation and hematopoietic differentiation, but BMP2 induced bone tissue formation activity was stronger. BMP4 had more effect on hematopoietic tissue, especially in adult. BMP2 induces ectopic bone formation and becomes an important inducing factor for bone tissue engineering: BMP4 can promote the proliferation of hematopoietic stem cells and induce hematopoietic stem cells to erythrocytes. The differentiation of granulocyte and macrophage cells can promote the regeneration and recovery of the damaged hematopoietic system. These characteristics make both BMP2 and BMP4 have good clinical application prospects. In the preparation of BMP, the eukaryotic expression system has the advantages of low expression level, difficult recovery of the expressed product, and high production cost. Prokaryotic expression system, especially Escherichia coli system, has the advantages of high expression efficiency, easy operation, easy monitoring, stable product, low production cost and so on. Therefore, we chose the E. coli expression system in the large-scale preparation of BMP. At present, the large-scale preparation of recombinant human bone morphogenetic protein 2 has been reported, the purpose of this study. It is to use the process of preparing BMP2, which is already mature at present, to prepare BMP4, to verify whether the two proteins can be prepared in the same fermentation process and similar purification methods.
