【摘要】：目的: 探索通過胸腺內(nèi)注射異基因骨髓細胞加腹腔注射環(huán)磷酰胺,以小鼠皮膚移植為模型,誘導(dǎo)移植免疫耐受,為免疫耐受提供理論和實驗依據(jù)。 方法: 提取供體C57BL/6(B6)小鼠(H-2~b)骨髓細胞,注入受體BALB/C(H-2~d)小鼠胸腺內(nèi),48h后腹腔注射200mg/kg的環(huán)磷酰胺,2w后移植供體C57BL/6(B6)小鼠(H-2b)和無關(guān)供體C3H(H-2~k)小鼠皮膚。共設(shè)空白對照組、CP對照組、BM對照組、實驗組、第三方供體組5個組。觀察皮膚存活時間;皮膚移植2w后對受體BALB/C(H-2~d)小鼠作單向混合淋巴細胞反應(yīng)(mixed lymphocyte reaction,MLR)、脾細胞增值反應(yīng)(Spleen cells proliferation reaction)及遲發(fā)型超敏反應(yīng)(delayed type hypersensitivity,DTH)耐受狀態(tài)的檢測。 結(jié)果: 耐受BALB/C小鼠獲得了對供體B6小鼠的特異性皮膚移植耐受,實驗組移植皮膚平均存活時間(MST)為22.00±2.51天,對照組MST為8.75±1.04天,耐受的BALB/C小鼠對第三供體C3H小鼠的移植皮膚仍正常排斥,MST為10.25±1.91天。單向混合淋巴細胞反應(yīng)和遲發(fā)型超敏反應(yīng)證明耐受的BALB/C小鼠對B6小鼠的脾細胞的反應(yīng)性特異性降低,而仍保持對無關(guān)第三者供體C3H小鼠的免疫反應(yīng)。耐受的BALB/C小鼠脾細胞對絲裂原(ConA)的增殖反應(yīng)在正常范圍。 結(jié)論: 使用胸腺內(nèi)注射供體異基因骨髓細胞,結(jié)合腹腔應(yīng)用環(huán)磷酰胺的
[Abstract]:Objective: to explore the induction of allogeneic immune tolerance by intrathymic injection of allogeneic bone marrow cells and intraperitoneal injection of cyclophosphamide in mice, and to provide theoretical and experimental evidence for immune tolerance. Methods: bone marrow cells were extracted from donor C57BL / 6 B6) mice and injected into the thymus of the recipient BALB / CnH-2B) mice. After 48 hours, 200 mg / kg cyclophosphamide was injected intraperitoneally. After 2 weeks, the donor C57BL / 6 B6 mice were transplanted into the skin of C57BL / 6B6) and the unrelated donor C3H2H 2) mice. The control group was divided into two groups: the CP control group and the BM control group, the experimental group were divided into two groups: the control group and the control group. But the immune response to C3H mice, which was not related to the third party donor, was maintained. The proliferative response of spleen cells to mitogen Cona in tolerant BALB/C mice was in the normal range.
