福建地區(qū)HCV基因型及Core蛋白的差異研究

【摘要】: 丙型肝炎病毒為單股正鏈RNA病毒,基因組全長約9.4kb。丙型肝炎進(jìn)展迅速且后果嚴(yán)重,極易轉(zhuǎn)變?yōu)楦斡不透伟?是危害人類健康的重要傳染病之一。HCV分為6個(gè)基因型,各型的地域分布,疾病表現(xiàn)以及對干擾素的治療反應(yīng)有很大差別,對HCV基因型及致病相關(guān)基因的結(jié)構(gòu)和功能的差別研究有助于判斷疾病的預(yù)后以及選擇合適的治療方案。本研究旨在了解福建地區(qū)HCV基因型,并對Core蛋白(HCV最重要的致病蛋白之一)進(jìn)行氨基酸差異分析。 本研究的第一部分為:巢式RT-PCR擴(kuò)增血清樣品HCV core基因部分區(qū)段(-12nt +343nt),克隆并測序;以Clustalw軟件對所克隆片段進(jìn)行系統(tǒng)進(jìn)化樹分析并分型,了解福建地區(qū)的HCV基因型分布。結(jié)果表明在102份福建地區(qū)HCV抗體陽性血清中,HCV檢出率為41.2%(共42份),系統(tǒng)進(jìn)化樹分析提示共有1b、2a、3b、6a四個(gè)不同的基因型,以6a和1b基因型為主。 本研究的第二部分為:RT-PCR擴(kuò)增并克隆上述四個(gè)基因型的HCV全長core基因并比較各基因型Core蛋白的氨基酸差別。結(jié)果顯示各型HCV Core蛋白共有33個(gè)氨基酸差別,主要集中4aa-21aa,68aa-78aa,106aa-121aa,139aa-163aa,182aa-191aa五個(gè)區(qū)域,不同基因型HCV在這些區(qū)域氨基酸的差別可能會導(dǎo)致Core蛋白的功能差異并影響其致病性。
[Abstract]:Hepatitis C virus (HCV) is a single-stranded positive strand RNA virus with a genome length of about 9.4 kb.Hepatitis C develops rapidly and has serious consequences. It is easy to be transformed into liver cirrhosis and liver cancer. HCV is one of the most important infectious diseases that endanger human health. HCV is divided into 6 genotypes. The regional distribution of each type, disease manifestation and therapeutic response to interferon are very different. The study on the structure and function of HCV genotypes and pathogenicity related genes is helpful to determine the prognosis of the disease and to select the appropriate treatment plan. This study aims to understand the genotype of HCV in Fujian province. Amino acid difference analysis was carried out for one of the most important pathogenic proteins of Core protein. The first part of this study was: nested RT-PCR amplification of the partial region of HCV core gene from serum samples, cloning and sequencing; The phylogenetic tree analysis and typing of the cloned fragments were carried out with Clustalw software to understand the distribution of HCV genotypes in Fujian. The results showed that 102 sera were positive for HCV antibodies in Fujian. The detection rate of HCV was 41.2%. Phylogenetic tree analysis showed that there were four different genotypes (6a and 1b). The second part of this study is as follows:. The HCV full-length core gene of the four genotypes was amplified and cloned by RT-PCR, and the amino acid differences of the Core proteins of the four genotypes were compared. There are 33 amino acid differences in Core protein. It is mainly concentrated in the five regions of 4aa-21aan 68aa-78aan 106aa-121aan 139aa-163aanjiao 182aa-191aa. The difference of amino acids between different genotypes of HCV in these regions may lead to the functional difference of Core protein and affect its pathogenicity.
