【摘要】：[目的]研究不同劑量及注射方案下未成熟樹突狀細(xì)胞(immature dendritic cell，，imDC)對(duì)大鼠同種異體器官移植免疫耐受的影響，探討未成熟樹突狀細(xì)胞誘導(dǎo)免疫耐受的機(jī)制。 [方法]聯(lián)合應(yīng)用低劑量重組大鼠GM-CSF(1.5ng／ml)和IL-4(4ng／ml)誘導(dǎo)Wistar大鼠骨髓細(xì)胞，培養(yǎng)出大量未成熟樹突狀細(xì)胞，利用形態(tài)學(xué)及FCM鑒定其成熟度。將SD大鼠分為4組：第一組為對(duì)照組，術(shù)前一天尾靜脈注射生理鹽水0.5ml，第二組為低劑量組，術(shù)前1天尾靜脈注射imDC 2×10~6，第三組為高劑量組，術(shù)前一天注射imDC 2×10~7，第四組為多次注射組，術(shù)前7天、術(shù)前1天、術(shù)后1天、術(shù)后5天注射imDC 2×10~6。以Wistar大鼠為供體，SD大鼠為受體，行頸部心臟移植，觀察移植心臟存活時(shí)間、術(shù)后移植后大鼠與供體大鼠及第三品系大鼠單向MLR、心肌病理及血清IL-2、IL-4、IL-10、IFN-r的水平變化。 [結(jié)果]1、新分離的細(xì)胞呈圓形，胞體小，在培養(yǎng)液中懸浮生長。培養(yǎng)至第3天，可見部分細(xì)胞附壁生長，細(xì)胞體積增大；在第5天出現(xiàn)毛刺狀細(xì)胞，但是毛刺短，分支少；至第7天，樹突狀細(xì)胞毛刺狀和聚集體仍不明顯。FCM示CD80、CD86均為低表達(dá)。 2、52例大鼠頸部心臟移植模型，手術(shù)成功率83.6％。取心時(shí)間(打開胸腔至心臟取出的時(shí)間)400.5±32.7秒，血管吻合時(shí)間2105.4±115.9秒。無藥物干預(yù)組，供心存活時(shí)間為6.8±1.0天。術(shù)中失敗11例，其中吻合口出血5例，血栓形成3例，受體呼吸停止2例，供心缺血時(shí)間過長1例；術(shù)后死亡6例，其中感染4例，不明原因死亡2例。 3、高劑量組移植心臟存活時(shí)間與對(duì)照組、低劑量組相比較，差別有統(tǒng)計(jì)學(xué)意義。多次注射組移植心臟存活時(shí)間與對(duì)照組、低劑量組相比較，差別有統(tǒng)計(jì)學(xué)意義。低劑量組與對(duì)照組移植心臟存活時(shí)間相比較，差別有統(tǒng)計(jì)學(xué)意義。高劑量組與多次注射移植心臟存活時(shí)間相比較，差別無統(tǒng)計(jì)學(xué)意義。 4、供體大鼠脾細(xì)胞作為刺激細(xì)胞時(shí)，高劑量組大鼠脾細(xì)胞SI值(3.28)，低劑量組SI值(3.59)，多次注射組SI值(3.71)，均明顯低于對(duì)照組(6.83)，差異有統(tǒng)計(jì)學(xué)意義(P＜0.01)，實(shí)驗(yàn)組間SI值無明顯差異。第三品系Lewis大鼠脾細(xì)胞作為刺激細(xì)胞時(shí)，高劑量組SI值(6.37)，低劑量組SI值(6.67)，多次注射組SI值(6.71)
[Abstract]:Objective To study the effects of immature dendritic cells ( imDC ) on immune tolerance in rat allogeneic organ transplantation under different dosage and injection protocols , and to explore the mechanism of immature dendritic cells to induce immune tolerance . Methods : Rat bone marrow cells were induced with low dose recombinant GM - CSF ( 1.5ng / ml ) and IL - 4 ( 4ng / ml ) . Results : 1 . The newly isolated cells were round , cell bodies were small , and the cells were suspended and grown in the culture medium . On the 3rd day , it was found that the cells were attached to the wall and the volume of the cells increased . On the 5th day , there was a burr - like cell , but the burr was short and the branches were few . On the 7th day , the burr - like and the aggregates of the dendritic cells were still not obvious . 2 銆
