【摘要】：天然自身抗體(natural autoantibody,NAA)可以定義為在沒(méi)有任何抗原免疫的情況下,正常機(jī)體產(chǎn)生的針對(duì)一種或多種自身抗原的抗體。NAA是構(gòu)成天然免疫的重要組成部分,可以維持自身穩(wěn)定、防止自身免疫病發(fā)生,具有抗炎、抗腫瘤監(jiān)視作用等。但由于相關(guān)研究進(jìn)展及實(shí)驗(yàn)方法的限制,對(duì)NAA的產(chǎn)生機(jī)制、產(chǎn)生NAA的自身反應(yīng)性B細(xì)胞的選擇/耐受機(jī)理等方面尚知之甚少。 抗體轉(zhuǎn)基因小鼠的應(yīng)用,為深入研究NAA產(chǎn)生機(jī)制提供了理想的平臺(tái)。目前已建立的多種自身抗體轉(zhuǎn)基因小鼠,從不同側(cè)面揭示了自身耐受的部分機(jī)制,初步探討了NAA的產(chǎn)生機(jī)理。本研究組對(duì)NAA家族之一的天然抗角蛋白自身抗體(anti-keratin autoantibody,AK auto Ab)進(jìn)行了多年深入、細(xì)致的研究,探討了AK auto Ab的許多重要功能。在此基礎(chǔ)上,應(yīng)用未免疫小鼠B細(xì)胞與骨髓瘤細(xì)胞融合,我們成功篩選到了分泌抗角蛋白抗體的雜交瘤,由未免疫小鼠獲得的AK
[Abstract]:Natural autobody NAA) can be defined as immune without any antigen. As an important part of innate immunity, NAA produced by normal body against one or more autoantigens can maintain self-stability, prevent autoimmune diseases, and have anti-inflammation. However, the mechanism of NAA production and the selection / tolerance mechanism of NAA autoreactive B cells are not well understood due to the progress of related research and the limitation of experimental methods. The application of antibody transgenic mice provides an ideal platform for further study on the mechanism of NAA production. A variety of autoantibody transgenic mice have been established at present. Some mechanisms of self tolerance were revealed from different aspects. The production mechanism of NAA was preliminarily discussed in this study. This study aimed at anti-keratin autoantibody, one of the natural anti-keratin autoantibodies of NAA family. AK auto Ab) has been studied deeply for many years, and many important functions of AK auto Ab have been discussed. Using the fusion of B cells and myeloma cells of unimmunized mice, we successfully selected hybridomas secreting anti-keratin antibodies, and obtained AK from unimmunized mice.
