【摘要】:【目的】:對(duì)脂肪來源的具有干細(xì)胞潛能的細(xì)胞進(jìn)行體外培養(yǎng),并對(duì)此類細(xì)胞體外生長的特性進(jìn)行研究,為進(jìn)一步的研究做基礎(chǔ)。 【材料與方法】:利用整形外科成熟的負(fù)壓吸脂術(shù)獲取脂肪組織,經(jīng)過一定處理后以酶解法得到具有增殖分化能力的幼稚細(xì)胞,此類細(xì)胞被稱為PLA(Processed Lipoaspirate)細(xì)胞,而后在體外培養(yǎng)擴(kuò)增。利用β-半乳糖苷酶染色法測(cè)定PLA的衰老率,利用流式細(xì)胞儀分析培養(yǎng)細(xì)胞的表面標(biāo)記物;一部分細(xì)胞放置于半固體培養(yǎng)基中培養(yǎng),利用油紅染色檢測(cè)細(xì)胞類型。 【結(jié)果】:在脂肪組織中確實(shí)存在能不斷增殖的未成熟細(xì)胞,多次傳代后仍保留增殖能力,第3代細(xì)胞的衰老率為:0.357±0.0178,而第15代細(xì)胞的衰老率為:0.371+0.0240(p0.05,無明顯差異),形態(tài)學(xué)及表面標(biāo)記物均與骨髓間充質(zhì)干細(xì)胞(Mesenchymal stem cell, MSC)類似,通過流式細(xì)胞技術(shù)(Fluorescence-activated cell sorter, FACS)鑒定,PLA細(xì)胞表面黏附分子CD44、CD29均高表達(dá),而造血干細(xì)胞調(diào)節(jié)因子CD34、CD45均低表達(dá)。半固體凝膠培養(yǎng)未能促進(jìn)組織分化。 【結(jié)論】:脂肪組織中確實(shí)存在一群具有極強(qiáng)增殖潛能的細(xì)胞,在多次分裂后,仍保持其增殖潛能。同時(shí)此類原始細(xì)胞的細(xì)胞表型與骨髓
[Abstract]:Objective: to culture adipose cells with stem cell potential in vitro and to study the characteristics of their growth in vitro. [materials and methods]: mature vacuum liposuction was used to obtain adipose tissue. After certain treatment, immature cells with proliferative and differentiation ability were obtained by enzymatic hydrolysis. These cells were called PLA(Processed lipoaspirate cells and were cultured and amplified in vitro. The senescence rate of PLA was determined by 尾 -galactosidase staining. The surface markers of cultured cells were analyzed by flow cytometry. Some of the cells were cultured in semisolid medium and the cell types were detected by oil red staining. [results]: there were immature cells in adipose tissue which could proliferate continuously, and the ability of proliferation remained after multiple passages. The senescence rate of the third passage cells was: 0.357 鹵0.0178. However, the senescence rate of the 15th passage cells was 0. 371 0. 0240 (p 0. 05). Morphological and surface markers were similar to mesenchymal stem cells (MSCs). The surface adhesion molecule (CD44) of PLA cells was identified by flow cytometry (FACSs) and fluorescence-activated cell sorter (FACSs). CD29 was highly expressed, while CD34 / CD45, a hematopoietic stem cell regulator, was low. Semisolid gel culture did not promote tissue differentiation. [conclusion]: there is indeed a group of cells with strong proliferative potential in adipose tissue, which remain proliferative potential after multiple division. Meanwhile, the phenotypes and bone marrow of these primitive cells are also present.
