【摘要】:細(xì)胞膜是細(xì)胞外大分子進(jìn)入細(xì)胞的主要屏障。電穿孔是指在瞬態(tài)電場(chǎng)作用下,瞬時(shí)增強(qiáng)細(xì)胞膜的通透性,在細(xì)胞膜的雙脂層上形成瞬時(shí)可逆微孔的生物物理現(xiàn)象。由此發(fā)展起來(lái)的電穿孔技術(shù)在DNA轉(zhuǎn)染、蛋白質(zhì)導(dǎo)入、細(xì)胞電融合等方面都有良好應(yīng)用。本文將其用于對(duì)離體細(xì)胞及惡性腫瘤的治療中,對(duì)腫瘤注射化療藥物并施加電脈沖,通過(guò)腫瘤生長(zhǎng)情況的檢測(cè),發(fā)現(xiàn)電脈沖結(jié)合化療藥物的電化學(xué)療法可以極大地改善腫瘤治療的效果。 生物組織的介電常數(shù)是一個(gè)重要的物理量,它反映了組織在外場(chǎng)作用下的極化情況。在實(shí)際的病理診斷和臨床治療中,可以根據(jù)組織和器官的介質(zhì)特性的變化和差異對(duì)病變組織進(jìn)行診斷和分析,在腫瘤及其它病癥的臨床診斷和治療中具有一定的應(yīng)用前景。通常生物組織的取樣數(shù)量十分有限,因此微量介質(zhì)的介電常數(shù)測(cè)量在生物介質(zhì)特性測(cè)量領(lǐng)域顯得十分必要。諧振腔微擾法特別適合對(duì)微小體積樣品的測(cè)試,很好解決了生物組織樣品取樣數(shù)量有限的問(wèn)題。 本文較詳細(xì)地分析了諧振腔微擾法,并由諧振腔微擾理論出發(fā),得到介質(zhì)復(fù)介電常數(shù)計(jì)算式。針對(duì)生物組織高含水量、高介電常數(shù)、高損耗的特性,采取了加大諧振腔體積、減小樣品體積及提高對(duì)樣品體積測(cè)試精度等措施,以提高生物樣品介電常數(shù)的測(cè)試精度。通過(guò)對(duì)水和不同濃度NaCl溶液介電常數(shù)的測(cè)試,驗(yàn)證了本系統(tǒng)的測(cè)量精度較高。本文首次對(duì)電脈沖處理下的腫瘤細(xì)胞懸液、小鼠血液的復(fù)介電常數(shù)進(jìn)行了測(cè)量,對(duì)電磁場(chǎng)生物效應(yīng)的研究具有一定參考價(jià)值,對(duì)組織介質(zhì)特性在臨床上的檢測(cè)有一定前瞻性的意義。
[Abstract]:The cell membrane is the main barrier of extracellular macromolecules into cells. Electroporation refers to the transient electric field, instantaneous increase of cell membrane permeability, biophysical phenomenon of transient reversible micropores in the bilayer of the cell membrane. The electroporation technique developed in the DNA transfected into protein, cell electrofusion etc. have a good application. This paper will be used in the treatment of malignant tumor cells in vitro and in tumor chemotherapy and applying electric pulses, detection of tumor growth through, found that electric pulse combined with electrical chemical therapy chemotherapy can greatly improve the effects of cancer treatment.
The permittivity of biological tissues is an important physical quantity, it reflects the polarization organization under the action of external field. The pathological diagnosis and clinical treatment of the actual, can the diagnosis and analysis of lesions according to the change of dielectric properties of tissues and organs and differences, has a certain application prospect in clinical diagnosis and for the treatment of tumors and other diseases. Usually very limited sampling quantity of biological tissues, so the trace of dielectric constant measurement in biological field measurement of dielectric properties is very necessary. Particularly suitable for testing small volume sample cavity perturbation method, solves the problem of biological tissue samples of a limited number of sampling.
This thesis makes a detailed analysis of the resonant cavity perturbation method, and based on perturbation theory, obtained the complex dielectric constant formula. According to the biological tissue high moisture content, high dielectric constant, high loss characteristics, to increase the cavity volume, reduce the sample volume and improve the testing precision of sample volume such measures, in order to improve the testing precision of biological samples. Based on the dielectric constant of water and different concentrations of NaCl solution dielectric constant test, verify the measurement precision of the system. This is the first research on the electric pulse treatment of tumor cell suspension, the complex permittivity of mice blood were measured with the reference value to the study of biological effect of electromagnetic field, has certain forward-looking significance to detection of tissue dielectric properties in clinic.
