【摘要】: 目的:建立實驗性神經(jīng)癥大鼠模型,觀察大鼠行為及中縫背核5-HT含量的變化規(guī)律,探討電針對實驗性神經(jīng)癥大鼠行為及中縫背核5-HT的影響。方法:健康成年SD大鼠50只,隨機分為正常對照組、避暗實驗組、基底節(jié)損毀組、電針治療一組、電針治療二組。5組動物均于兩周后進行行為測試,測試后處死,常規(guī)灌注冰凍切片,采用SABC免疫組化和計算機圖像分析技術(shù),檢測中縫背核5-HT神經(jīng)元陽性細胞灰度值。結(jié)果:造模后,兩組模型大鼠水平運動次數(shù)和垂直運動次數(shù)均明顯降低,中縫背核內(nèi)5-HT水平較正常組減低(P<0.05),電針干預(yù)后,治療組5-HT含量較模型組有所增加。結(jié)論:1.避暗實驗和基底節(jié)損毀法可成功建立神經(jīng)癥大鼠模型。2.模型組大鼠腦干中縫背核5-HT含量降低,與中醫(yī)肝郁證有著共同的病理基礎(chǔ),均與體內(nèi)單胺類神經(jīng)遞質(zhì)及其生理活性相關(guān)。3.電針百會、印堂穴可使模型大鼠活動增多,中樞5-HT趨于正常,證明了電針百會、印堂可以改善模型動物的病理狀態(tài),從治療手段有效性的反證角度,對兩種模型的建立與評價提供了有力的支持。 前言 隨著現(xiàn)代社會工作和生活節(jié)奏不斷加快,競爭等應(yīng)激事件日益增加,人們承受的心理壓力也日益增大。無論是發(fā)達國家還是發(fā)展中的中國,心理健康問題越來越嚴重,其中又以神經(jīng)癥的發(fā)病率上升最為明顯。神經(jīng)癥(neuroses),舊稱神經(jīng)官能癥,是臨床醫(yī)學(xué)中的重要疾病之一。本病為一組精神障礙,主要表現(xiàn)為:精神活動能力低下、煩惱緊張、焦慮抑郁、恐怖、強迫癥狀、疑病癥狀、分離癥狀、轉(zhuǎn)換癥狀或各種軀體不適感。系統(tǒng)地研究神經(jīng)癥的生物學(xué)基礎(chǔ),有助于對神經(jīng)癥開展有針對性的預(yù)防和治療。然而,由于對人類活體研究存在技術(shù)上和倫理上的限制,目前對本病的生物學(xué)機制仍知之甚少。因此,建立一個合理的動物模型實屬必要。 神經(jīng)癥當屬祖國醫(yī)學(xué)中“郁癥”范疇。針灸治療本類疾病在我國已有悠久的歷史,早在《內(nèi)經(jīng)》時代就有針灸治療本類疾病的記載,在歷代文獻記載中,約有130個穴位與治療精神疾病有關(guān),,如四關(guān)穴、十三鬼穴等。當今關(guān)于針灸治療抑郁癥、強迫癥、焦慮癥等神經(jīng)癥的報道更是不勝枚舉。諸多研究顯示:針灸治療神經(jīng)癥的效果與臨床一線藥物相當,且具有無毒副作用、經(jīng)濟便捷的優(yōu)勢。 本論文既以此為出發(fā)點,通過避暗實驗和基底節(jié)損毀實驗建立神經(jīng)癥大鼠模型,并采用反證法探討電針治療本病的作用機理,對模型的建立與評價提供有力支持。
[Abstract]:Objective: to establish a rat model of experimental neurosis and observe the changes of behavior and 5-HT content in dorsal raphe nucleus. To investigate the effect of electroacupuncture on the behavior of experimental neurosis rats and 5-HT in dorsal raphe nucleus. Methods: 50 healthy adult SD rats were randomly divided into normal control group, dark avoidance group and basal ganglion lesion group. The animals of the first group of electroacupuncture treatment and the 2 groups of electroacupuncture treatment group were given behavioral tests two weeks later. After the test, the animals were killed, frozen sections were infused routinely, and SABC immunohistochemistry and computer image analysis technique were used. Results: after the establishment of the model, the number of horizontal and vertical movements of the two groups were significantly decreased. The level of 5-HT in dorsal raphe nucleus was lower than that in normal group (P < 0.05). The 5-HT content in the treatment group was higher than that in the model group. Conclusion the neurotic rat model of neurosis can be established successfully by avoiding dark experiment and basal ganglia lesion. The content of 5-HT in the dorsal raphe nucleus of the brain stem of the model group is decreased. There is a common pathological basis with traditional Chinese medicine liver depression syndrome, which is related to monoamine neurotransmitters and their physiological activities. 3. EA Baihui, Yantang point can increase the activity of model rats, and the central 5-HT tends to normal. It is proved that electroacupuncture and Yantang can improve the pathological state of the model animals. From the point of view of the effectiveness of the therapeutic methods, it provides a strong support for the establishment and evaluation of the two models. Mental health problems are becoming more and more serious, in which the incidence of neurosis is the most obvious. Neurosis, formerly known as neurosis. This disease is a group of mental disorders, mainly manifested in: low mental activity, anxiety and depression, anxiety and depression, phobia, compulsive symptoms, hypochondriac symptoms, separation symptoms. A systematic study of the biological basis of neurosis is helpful for targeted prevention and treatment of neurosis. Due to the technical and ethical limitations of human living research, little is known about the biological mechanism of the disease, so it is necessary to establish a reasonable animal model. Neurosis belongs to the category of "depression" in Chinese medicine. Acupuncture and moxibustion treatment of this kind of disease has a long history in our country, as early as in the "Internal Classic" era, acupuncture and moxibustion treatment of this kind of disease has been recorded in the literature records of the past dynasties. There are about 130 acupoints related to the treatment of mental illness, such as Siguan point, 13 ghost points, etc. Today about acupuncture treatment of depression, obsessive-compulsive disorder. There are numerous reports of anxiety disorders and other neuroses. Many studies show that acupuncture and moxibustion treatment of neurosis is as effective as clinical first-line drugs and has the advantages of no toxic side effects and convenient economy. This paper not only takes this as the starting point, through the dark avoidance experiment and the basal ganglia damage experiment to establish the neurosis rat model, and uses the counter-syndrome method to explore the mechanism of electroacupuncture treatment of this disease. Support the establishment and evaluation of the model.
