【摘要】： 心肌肌鈣蛋白是心肌運(yùn)動(dòng)的調(diào)節(jié)蛋白,心肌梗死等損傷性疾病發(fā)生時(shí),其亞單位肌鈣蛋白T和肌鈣蛋白I釋放入血,是診斷心肌損傷的特異性標(biāo)志物。本研究通過(guò)組織勻漿、蛋白層析技術(shù)實(shí)現(xiàn)對(duì)牛心肌肌鈣蛋白T的提取和純化,通過(guò)SDS-PAGE和ELISA方法鑒定獲得蛋白的性質(zhì)。獲取的蛋白免疫動(dòng)物后,采用經(jīng)典的細(xì)胞融合技術(shù),制備雜交瘤細(xì)胞,通過(guò)ELISA方法鑒定培養(yǎng)細(xì)胞上清,經(jīng)雜交瘤細(xì)胞克隆,分選,擴(kuò)增并穩(wěn)定凍存,獲得5株陽(yáng)性細(xì)胞株,為心肌肌鈣蛋白檢測(cè)試劑的國(guó)產(chǎn)化奠定實(shí)驗(yàn)基礎(chǔ)。
[Abstract]:Cardiac troponin is the regulating protein of myocardial movement. When myocardial infarction and other injury diseases occur, its subunit troponin T and troponin I release into the blood. In this study, bovine cardiac troponin T was extracted and purified by tissue homogenate and protein chromatography. The obtained protein was immunized by SDS-PAGE and ELISA. The hybridoma cells were prepared by classical cell fusion technique. The supernatants of cultured cells were identified by ELISA method, cloned, sorted, amplified and frozen stably by hybridoma cells, and 5 positive cell lines were obtained, which laid the experimental foundation for the localization of cardiac troponin detection reagent.
