【摘要】: 實驗?zāi)康? 觀察在體外培養(yǎng)條件下沖擊波誘導(dǎo)健康成年人骨髓間充質(zhì)干細(xì)胞(hMSCs)向成骨細(xì)胞分化特點,研究適宜沖擊波作用強度,并探討其機制。 實驗方法: 1、在體外對健康成年人的骨髓間充質(zhì)干細(xì)胞進(jìn)行分離、傳代培養(yǎng),經(jīng)免疫組織化學(xué)方法鑒定后,分別采用不同強度的低能沖擊波進(jìn)行誘導(dǎo),觀察細(xì)胞變化并進(jìn)行檢測,確定最佳沖擊波誘導(dǎo)強度。 2、對鑒定后的骨髓間充質(zhì)干細(xì)胞,采用最佳強度的沖擊波進(jìn)行誘導(dǎo),并在不同的時間進(jìn)行RT-PCR檢測,觀察c-fos和c-jun基因表達(dá)的變化。 實驗結(jié)果: 沖擊波誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向成骨細(xì)胞分化的最佳作用強度為8.5kV,120次,成骨作用明顯優(yōu)于對照組(P0.01)。采用最佳強度沖擊波作用后,c-fos和c-jun基因表達(dá)量較對照組明顯增加,45min達(dá)到高峰。 實驗結(jié)論: 低能沖擊波在適宜條件下可誘導(dǎo)骨髓間充質(zhì)干細(xì)胞向成骨細(xì)胞分化,c-fos和c-jun基因表達(dá)增加。
[Abstract]:Objective: Objective: to observe the differentiation of bone marrow mesenchymal stem cells (hMSCs) into osteoblasts induced by shock wave in vitro, to study the suitable shock wave intensity, and to explore its mechanism. Experimental methods: 1. Bone marrow mesenchymal stem cells from healthy adults were isolated and cultured in vitro. The changes of cells were observed and detected to determine the optimal intensity of shock wave induction. 2. The bone marrow mesenchymal stem cells were induced by shock wave with the best intensity, and RT-PCR was detected at different time. The changes of c-fos and c-jun gene expression were observed. Experimental results: The best effect of shock wave on differentiation of bone marrow mesenchymal stem cells into osteoblasts was 8.5 kV / 120 times. The expression of c-fos and c-jun genes was significantly increased to a peak at 45 minutes after the best shock wave was used in the osteogenesis group, and the expression of c-fos and c-jun genes in the control group was significantly higher than that in the control group. The experimental conclusions are as follows: Low energy shock wave could induce the expression of c-fos and c-jun genes in osteoblast differentiation of bone marrow mesenchymal stem cells under suitable conditions.
