人胎兒卵巢體外培養(yǎng)的實(shí)驗(yàn)研究

【摘要】:目的:探討人胎兒卵巢在體外培養(yǎng)及冷凍保存復(fù)蘇后不同條件下卵泡發(fā)育并分泌雌激素情況,探討體外培養(yǎng)條件下卵泡刺激素(follicle stimulating hormone,FSH)、N-乙酰-L-半胱氨酸(N-acetyl-L-cysteine,NAC)對(duì)人胎兒卵巢發(fā)育的影響;了解人胎兒卵巢組織的最佳體外培養(yǎng)條件,為后續(xù)研究提供必要的實(shí)驗(yàn)依據(jù)。 方法:選取20~28周的胎兒卵巢30例采用器官組織培養(yǎng)的方法進(jìn)行體外培養(yǎng),按照培養(yǎng)液不同成分分為O組(O液:生理鹽水)、A組(A液:生理鹽水加20%新生牛血清)、B組(B液:基本培養(yǎng)液)、C組(C液:含0.1mmol/l NAC的基本培養(yǎng)液)、D組(D液:含0.5u/ml FSH的基本培養(yǎng)液)、E組(E液:含0.1mmol/l NAC、0.5u/ml FSH的基本培養(yǎng)液)每3天更換培養(yǎng)液并保存培養(yǎng)液以液相平衡競(jìng)爭(zhēng)放射免疫分析法(RIA)進(jìn)行雌二醇(estradiol,E_2)測(cè)定,并留取培養(yǎng)后人胎兒卵巢組織塊進(jìn)行組織學(xué)觀察。 結(jié)果:1.相同培養(yǎng)時(shí)間下,B組卵巢雌二醇分泌量高于O組、A組,二者相比差異顯著(P0.05);O組與A組,培養(yǎng)后第4~6天、7~9天卵巢雌二醇分泌量低于第1~3天卵巢雌二醇分泌量,二者差異顯著(P0.05);B組培養(yǎng)第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天分泌量,二者相比差異顯著(P0.05)。2.相同培養(yǎng)時(shí)間下,新鮮組與凍融組之間,卵巢雌二醇分泌量無(wú)顯著性差異(P0.05),相同培養(yǎng)時(shí)間下,新鮮組與凍融組之間,卵巢各期卵泡構(gòu)成比無(wú)顯著性差異(P0.05);而新鮮組與凍融組內(nèi)培養(yǎng)第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天卵巢雌二醇分泌量,二者差異性顯著(P0.05)。新鮮組與凍融組內(nèi),培養(yǎng)第3天、6天、9天后卵巢中較為成熟的卵泡比例明顯高于未培養(yǎng)組(P0.05)。3.相同培養(yǎng)時(shí)間下,C組、D組、E組卵巢雌二醇分泌量高于B組,二者差異性顯著(P0.05)。C組、D組、E組內(nèi),培養(yǎng)第4~6天、7~9天卵巢雌二醇分泌量高于第1~3天卵巢雌二醇分泌量,二者差異性顯
[Abstract]:Objective: To explore the human fetal ovary in vitro and frozen thawed under different conditions of follicular development and the secretion of estrogen, to investigate the in vitro follicle stimulating hormone (follicle stimulating, hormone, FSH), N- acetyl -L- cysteine (N-acetyl-L-cysteine, NAC) on human fetal ovarian development; to understand the optimal culture conditions in vitro of human fetus ovarian tissue, provide the necessary experimental basis for further study.
Methods: the fetal ovary from 20 to 28 weeks, 30 cases with organ tissue culture method in vitro and in accordance with the different medium components were divided into O group (O solution: physiological saline), A group (A solution: normal saline and 20% newborn calf serum), B group (B solution: basic medium), group C (basic medium C liquid containing 0.1mmol/l NAC), D group (basic medium D liquid containing 0.5u/ml FSH), E group (basic medium E solution containing 0.1mmol/l NAC, 0.5u/ml: FSH) replaced the culture medium every 3 days and the preservation of culture liquid in liquid phase equilibrium competition radiation immunoassay (RIA) of estradiol (estradiol, E_2) were determined, and take the cultivation of human fetal ovarian tissue blocks for histological observation.
