【摘要】: 本研究建立了煙曲霉感染的動物模型,篩選出強(qiáng)、弱毒株,利用比較蛋白質(zhì)組學(xué)技術(shù),分析了兩者的蛋白質(zhì)表達(dá)譜,發(fā)現(xiàn)強(qiáng)、弱毒株之間蛋白質(zhì)組成存在差別,經(jīng)軟件分析匹配選取39個經(jīng)質(zhì)譜檢測,鑒定出6個蛋白質(zhì)。其中,強(qiáng)毒株中有2個表達(dá)上調(diào)的蛋白質(zhì)(Cat1P,AspF3)認(rèn)為與致病性相關(guān);相應(yīng)基因分析結(jié)果表明蛋白質(zhì)的這種表達(dá)差異主要是在轉(zhuǎn)錄水平發(fā)生的。在弱毒株中表達(dá)上調(diào)的蛋白質(zhì)有4個(actin,NADP linked glutamate dehydrogenase , guanine nucleotide-binding beta subunit liked protein,orotidine 5`-phosphate decarboxylase)分別涉及細(xì)胞活動、代謝合成及信號傳導(dǎo)途徑的某個環(huán)節(jié)。本研究還進(jìn)行了煙曲霉與念珠菌蛋白質(zhì)組的二維凝膠電泳圖譜比較,顯示一些主要高豐度表達(dá)蛋白質(zhì)不在同一區(qū)域內(nèi),認(rèn)為它們的蛋白質(zhì)組存在明顯差異。本實驗為煙曲霉致病機(jī)制的研究、抗真菌藥物及疫苗的研發(fā)提供了重要的理論基礎(chǔ)和實驗依據(jù)。
[Abstract]:In this study, the animal model of Aspergillus fumigatus infection was established, and the strong and weak strains were screened. The protein expression profiles of the two strains were analyzed by comparative proteomics technique, and the difference of protein composition between the strong and weak strains was found. By software analysis, 39 proteins were identified by mass spectrometry. Among them, 2 of the virulent strains expressed up-regulated protein Cat1PAspF3, which were thought to be related to pathogenicity. The results of gene analysis showed that the difference of protein expression was mainly at the transcriptional level, and four proteins were up-regulated in attenuated strains. NADP linked glutamate dehydrogenase. Guanine nucleotide-binding beta subunit liked protein. Orotidine 5 '-phosphate decarboxylase is involved in cell activity. This study also compared the proteome of Aspergillus fumigatus and candida by two-dimensional gel electrophoresis, which showed that some of the major high abundance proteins were not in the same region. This study provides an important theoretical basis and experimental basis for the study of pathogenic mechanism of Aspergillus fumigatus and the research and development of antifungal drugs and vaccines.
