【摘要】：糖尿病(diabetes mellitus,DM)中醫(yī)又稱為“消渴病”或“消癉”,是由胰島素合成與分泌減少,或胰島素受體及受體后功能障礙而引起的以高血糖為生化特征的一種常見內分泌代謝疾病。 小型豬體型小、遺傳性能穩(wěn)定、器官功能和組織結構與人體相似度大,具有雜食性,實驗環(huán)境和條件易于控制。在我國小型豬資源較為豐富,適于作醫(yī)學動物實驗的有五指山微型豬、環(huán)江香豬、巴馬小型豬、西雙版納小型豬、芷豬等品種/品系。廣西巴馬小型豬24項血液生理指標中有14項與人類相似,表明巴馬小型豬生理功能與人具有相似性。SPF小型豬的血液、尿液生理生化參考值變動幅度小,SPF小型豬血液成分變化趨勢與SPF大、小鼠血液成分變化趨勢一致。鏈脲佐菌素(STZ)能夠選擇性地破壞某些種屬動物胰島的β細胞,導致糖尿病的發(fā)生。四氧嘧啶(Alloxan)40mg/Kg體重腹腔注射,配以高糖飼料復制出糖尿病胃腸動力障礙模型。細胞凋亡在糖尿病并發(fā)癥的發(fā)生中發(fā)揮重要作用,在高血糖狀態(tài)下,抗凋亡基因Bcl-2表達降低,而促凋亡基因Bax表達增加,導致細胞凋亡數(shù)目增加。通過對細胞凋亡的調控機制的研究,可望找到延緩或逆轉細胞凋亡,防治糖尿病及其并發(fā)癥的方法。 中醫(yī)學認為糖尿病屬陰虛燥熱,以陰虛為本,以燥熱為標,與肺脾(胃)腎三臟相關。脾胃功能失調、脾氣虛弱是糖尿病的病機。有“脾虛病機論”、“治脾論”、“調理脾胃升降治糖尿病”等觀點。糖尿病性胃病或以癥狀命名,如“痞滿”、“噫氣”、“痰飲”;或據(jù)病機將本病診為“胃緩”,因中虛氣滯,通降無力而致。 資料表明,患者胃排空延遲,出現(xiàn)厭食、打呃、腹脹等臨床癥狀。中醫(yī)提出補氣健脾、和胃理氣,益氣養(yǎng)陰、行氣活血和醒脾運脾等治則治法,具有顯著臨床療效。糖尿病胃病屬本虛標實之證。治療時應予具有健脾理氣、活血化瘀適當加用益腎、養(yǎng)胃作用的方藥。中藥復方“脾胃康”是根據(jù)糖尿病胃病發(fā)病機制及中醫(yī)治則研制并使用的經驗方,臨床應用療效顯著。 為了探討建立小型豬糖尿病模型的程序和方法,進一步研究糖尿病并發(fā)癥的發(fā)病機制,揭示中藥復方“脾胃康”的藥物作用機理,特開展了本實驗研究。 本研究旨在通過對廣西巴馬小型豬的生理、生化指標及其在不同時期的生長變化、消化道組織的超微結構進行研究,探討巴馬小型豬的實驗動物化;建立小型豬胃電極埋植手術操作程序和胃電檢測方法,檢測小型豬的胃電參考數(shù)據(jù)及其變化規(guī)律;對小型豬進行糖尿病模型復制試驗,探討初步建立小型豬糖尿病模型的復制方法;觀測糖尿病大鼠胃組織的病理改變和凋亡相關基因Bax、Bcl-2蛋白表達變化,探討中藥復方“脾胃康”的作用機制。 巴馬小型豬3月齡時平均體重為2.87±0.58kg,體長34.29±4.35cm。12月齡時平均體重達到6.50±0.44kg,體長達到66.67±4.03cm。到18月齡平均體重和
[Abstract]:Diabetes (diabetes mellitus, DM) of traditional Chinese medicine is also called "diabetes" or "Xiaodan", is composed of insulin synthesis and secretion reduced, or dysfunction of the insulin receptor and post receptor induced by high glucose as the biochemical features of a kind of common endocrine and metabolic diseases.
Pigs are small, genetic stability, organ function and tissue structure with human similarity, is omnivorous, the experimental environment and the condition is easy to control. In the small pig resource in China is relatively rich, suitable for medical animal experiments with Five Fingers Group miniature pig, Huanjiang pig, Bama miniature pig, Xishuangbanna pig, pig Zhi varieties / lines. 24 blood physiological index of Guangxi Bama miniature pig in 14 and that is similar to that of human physiological function of Bama miniature pigs and people with similar.SPF mini pig's blood, urine, physiological and biochemical changes in small amplitude of the reference value, SPF mini pig blood composition change trend and SPF, changes of blood components in mice the same trend. Streptozotocin (STZ) can selectively destroy certain species of animal pancreatic beta cells, leading to the occurrence of diabetes. Four alloxan (Alloxan) 40mg/Kg intraperitoneal injection, with high sugar diet complex A model of diabetic gastrointestinal motility disorders. Apoptosis plays an important role in the occurrence of complications of diabetes, high blood sugar condition, anti apoptosis gene Bcl-2 expression decreased, while the expression of apoptotic gene Bax increased, resulting in cell apoptosis. The increase in the number of study on the mechanism of regulation on cell apoptosis, may be found to delay or reverse the apoptosis methods, prevention and treatment of diabetes and its complications.
Traditional Chinese medicine thinks that diabetes is Yin scorching, yin deficiency, the standard for heat, spleen (stomach) and lung kidney three dirty. Dysfunction of spleen and stomach, spleen qi deficiency is the pathogenesis of diabetes. "The pathogenesis theory", "Treat Spleen", "spleen and stomach and treat diabetes" concept. Diabetic gastropathy or in the name of symptoms, such as "fullness", "Yi Qi", "phlegm"; or according to the pathogenesis of the disease will be diagnosed as "stomach slow", because the virtual stagnation, Tongjiang and weakness.
Data show that the delayed gastric emptying in patients with anorexia, abdominal distension, hiccup, clinical symptoms of TCM. The spleen and stomach, Qi, tonifying qi and nourishing Yin, Qi and blood, spleen and spleen treatment, with significant clinical efficacy. Diabetic gastropathy is superficiality syndrome. Treatment should be with Spleen Qi. Appropriate with Yishen Huoxue Huayu Decoction, Yangwei effect. In order to explore the procedure and method of establishing miniature pig diabetes model, we further studied the pathogenesis of diabetic complications, and revealed the mechanism of Chinese herbal compound "spleen stomach Kang".
The purpose of this study is to through the physiology of Guangxi Bama mini pig, biochemical index and growth in different periods, to study the ultrastructure of gastrointestinal tissue, explore the Bama miniature pig experimental animal; establishment of miniature pig gastric electrode implantation operation procedure and gastric electrical detection method, detection of porcine gastric electrical reference data and changes; replicate test model of diabetes on mini pigs, replication method study of the establishment of diabetic model in pigs; pathological change and apoptosis related gene Bax in gastric tissue observed in diabetic rats, the expression changes of Bcl-2 protein, and to explore the mechanism of traditional Chinese medicine "piweikang".
The average body weight of Bama miniature pigs is 2.87 + 0.58kg, the body length is 34.29 + 4.35cm.12, the average body weight reaches 6.50 + 0.44kg at the age of the month, and the body length reaches 66.67 + 4.03cm. to 18 month old, the average body weight is 3 month old.
