【摘要】:經(jīng)典的formalin 痛模型即腳掌注射formalin 引起的持續(xù)約1 小時的雙相性痛行為反應(yīng)。由于能較好地模擬臨床急性炎癥痛及能采用定性和半定量的指標(biāo)對痛反應(yīng)進(jìn)行測量,因而被廣泛應(yīng)用于疼痛機(jī)制的研究。新近的研究顯示,formalin 皮下注射除引起上述急性炎性痛外,還可以引起持續(xù)性的痛敏狀態(tài),如持續(xù)約4 周的熱輻射痛敏反應(yīng)及同時觀察到的脊髓背角小膠質(zhì)細(xì)胞的激活。在以往的實驗中,本研究室及其他研究者的觀察都證實,NMDA 受體在formalin 引起的急性炎性痛中發(fā)揮重要作用。本實驗在外周注射formalin 的基礎(chǔ)上,通過痛行為觀察(熱輻射刺激縮足反射潛伏期)、對小膠質(zhì)細(xì)胞活化增殖的免疫組化檢測(OX-42 陽性表達(dá)),結(jié)合外周及鞘內(nèi)注射NMDA 受體拮抗劑MK-801,試圖證實外周與脊髓NMDA 受體在formalin 所致的長時程痛敏及脊髓小膠質(zhì)細(xì)胞的活化中發(fā)揮重要作用。 實驗在Sprague-Dawley 雄性大鼠(90~100g)進(jìn)行。本實驗分兩部分,第一部分是通過外周注入MK-801 觀察外周傳入感覺神經(jīng)末稍上NMDA 受體在formalin 引起的長時程痛敏反應(yīng)及脊髓小膠質(zhì)細(xì)胞活化中的作用。包括免疫組化實驗和行為學(xué)觀察。在免疫組化實驗中,根據(jù)給藥方法的不同,分為多次給藥組和單次給藥組(n=6)。多次給藥組中,在動物兩側(cè)腳掌注入formalin 前15 分鐘,將NMDA 受體的拮抗劑MK-801(分為三個劑量組即10 nmol、30 nmol、50 nmol)注入左側(cè)腳掌皮下,而在右側(cè)注入生理鹽水(NS),并且在formalin 注射后的第1 天、第2 天、第3 天、第4 天分別重復(fù)給予原劑量的MK-801或NS。在注射formalin后第7天進(jìn)行OX-42表達(dá)的免疫組化檢測,對比背角兩側(cè)小膠質(zhì)細(xì)胞的免疫組化反應(yīng),進(jìn)行圖像分析及統(tǒng)計學(xué)處理。單次給藥組也分為三個劑量組即50 nmol、100 nmol、150 nmol,與多次給藥組不同的是注射formalin 前15 分鐘一次全量給予MK-801 或NS,到第7 天進(jìn)行免疫組化檢測。在痛行為實驗觀察中,分為實驗組(30 nmol MK-801+formalin)和對照組(NS+formalin),采用多
[Abstract]:Diphasic pain behavior response formalin pain model is the classic peripheral injection of formalin induced lasted about 1 hours.
