猴免疫缺陷病毒SIVmac239感染中國恒河猴建立AIDS動物模型的研究

【摘要】:動物模型是研究許多疾病不可缺少的工具。因靈長類動物在進(jìn)化關(guān)系上與人類最為接近,所以非人靈長類動物已成為研究人類疾病十分重要的實驗動物。自從證實HIV是AIDS病因后,即開始尋找和研究有關(guān)合適感染HIV的動物模型。經(jīng)過大量實驗篩選,發(fā)現(xiàn)非人靈長類動物是較為理想的用于HIV發(fā)病機制、藥物治療和疫苗開發(fā)的動物模型,現(xiàn)已廣泛應(yīng)用于AIDS的研究領(lǐng)域。建立合適的動物模型已成為國家重大需求的基礎(chǔ)問題,,這將有助于我國對AIDS感染和發(fā)病機理的研究,推動抗AIDS新藥和疫苗的開發(fā),從而有利于AIDS的防制工作。 本論文研究中,首先采用分子克隆技術(shù)復(fù)建SIVmac239和SHIV89.6病毒株。然后用SIVmac239靜脈感染2只中國恒河猴,建立AIDS非人靈長類動物模型。攻毒后,定期對體重、外周血CD4~+T細(xì)胞數(shù)量、PBMC前病毒DNA、抗體、組織器官病理變化進(jìn)行檢測,共監(jiān)測121天。在感染早期,便可從宿主細(xì)胞中檢測到前病毒,并分離出具感染性的SIVmac239;同時,外周血CD4~+T淋巴細(xì)胞急劇下降,并一直維持在一個較低水平;對血漿抗體的檢測結(jié)果表明,動物機體產(chǎn)生了針對病毒的特異性體液免疫反應(yīng);在感染121天時,腸道、部分淋巴結(jié)等組織發(fā)生一定程度的病理變化,特別在小腸粘膜部位發(fā)生淋巴組織增生。這些結(jié)果說明,SIVmac239成功感染2只中國恒河猴,可應(yīng)用這種模型進(jìn)行AIDS相關(guān)研究工作。
[Abstract]:Animal models are powerful tools for many diseases. Because primate animal in evolutionary relationship with most similar to human, so non-human primate animal experimental animal studies of human disease has become very important. Since that HIV is the cause of AIDS, which began to look for and Research on appropriate animal models of HIV infection after a lot of experiments. Screening, found the non-human primate animal is an ideal animal model for the pathogenesis of HIV, drug therapy and vaccine development, research field and has been widely used in AIDS. To establish a suitable animal model has become the basic problem of the major needs of the country, which will be helpful to our study of AIDS infection and pathogenesis. To promote the development of anti AIDS drugs and vaccines, which is conducive to the prevention and control of AIDS.
In this paper, firstly by molecular cloning technique and rehabilitation SIVmac239 SHIV89.6 strains. Then using SIVmac239 2 China intravenous infection of monkeys in Ganges RIver, the establishment of non-human primate animal models of AIDS. When challenged on a regular basis weight, the number of peripheral blood CD4~+T cells, PBMC virus DNA antibody, the histopathological changes of detection, were monitored for 121 days. In the early stage of infection, the virus can be detected from the host cells, and isolated infection of SIVmac239; at the same time, peripheral blood CD4~+T lymphocytes decreased sharply, and has remained at a low level; the detection of plasma antibodies showed that the animal body to produce specific humoral immune response to virus infection; intestinal in 121 days, and the pathological changes in some lymph nodes tissue to a certain extent, especially in parts of intestinal mucosal lymphoid hyperplasia. These results suggest that SIV Mac239 has successfully infected 2 Chinese Ganges RIver monkeys. This model can be used for AIDS related research.
